Caffeine is odorless, bitter taste substance which can be naturally found in coffee, cocoa, tea leaves, and is intentionally added in food and pharmaceutical products. It can also be found in surface water in small concentrations where is often used as an excellent indicator of human waste. The aim of the work is determination of caffeine content in food, beverages, analgesics and surface water using solidphase extraction followed by high-performance liquid chromatography (HPLC). Caffeine content was determined in 12 commercial tea and coffee products, non-alcoholic energy drinks and food, 5 combined preparations of analgesics and the Danube samples collected from 7 representative locations. The results showed that caffeine content in food ranged 5,6-158 mg/100 g, tea samples 24,71-30,81 mg/100 ml, coffee samples 1328-3594 mg/100 g, energy drinks 9,69-30,79 mg/100 ml and in the Danube samples 15,91-306,12 ng/l. Caffeine content in combined commercial formulations of non-narcotic analgesics of all brands did meet specifications. The data suggested that the proposed HPLC method can be used for routine determination and control of caffeine content in different matrices.

Abstract:In modern times in all age groups energy supplements containing different amounts of Caffeine and Taurine are applied. Caffeine is purine alkaloid, which stimulates central nervous system action, enhances the strength and frequency of the cardiac contractions and increases the excretion of urine. Taurine is a sulfur containing amino acid, which possesses many fundamental biological roles including: effect on synaptic transmission in the central nervous system, cardiotropic action, antioxidant and anticonvulsant activity, improvement of energy processes, stimulation of reparative processes in tissues, protection of eyes cataract, decrease of cholesterol and stimulation of immune system. The combination of Caffeine and Taurine provide benefit due to obtaining synergism of pharmacological effects in increasing of physical activity, stimulation of brain action, cognition, memory and attention. In connection with the significant enlarging of the consumption of energy drinks, especially by children and young people in recent years the requirements for regulation and control of the labeling of these products in many countries are enlarged. In many food additives Caffeine and Taurine are added in not labeled high concentrations, which can provoke and increase their side effects. High consumption of Caffeine enhances its adverse effects on body: anxiety, headache, insomnia, nervousness, respiratory disorders, tachycardia, tremor, dehydration. In children the adverse reactions of Caffeine in much lower doses than adults are occurred. In high concentrations Taurine has adverse effects on brain activity and can induce psoriasis. The result of combination of Caffeine and Taurine is associated with increased diuretic effect and loss of water and salts from the body, especially in children and young people. Because of these facts the quality and quantity control of included compounds in food supplements is important for their health safety.

Novel insights into the strategy of highly precise, carbon‐based electrochemical sensors are presented by exploring the excellent properties of graphene oxide (GO) and multiwalled carbon nanotube composites (GO‐MWCNTs/CPE) for the sensitive determination of tramadol hydrochloride (TRH). Cyclic voltammetry, differential pulse voltammetry, chronoamperometry (CA), and electrochemical impedance spectroscopy (EIS) scanning electron microscopy, and X‐ray diffraction (XRD) techniques were used to characterize the properties of the sensor. The linear response obtained for TRH using the GO‐MWCNTs/CPE was found to be over the range of 2.0x10−9 to 1.1x10−3 M with a good linearity and high correlation (0.9996). The limits of detection and quantification were found to be 1.50x10−10 M and 4.99 x 10−10 M, respectively. The proposed sensor was applied for determination of TRH in the presence of presence of co‐formulated drugs ketorolac tromethamine (KTM) and paracetamol (PAR). The sensor was shown to successfully apply to the determination of TRH in plasma as real samples. Satisfactory recoveries of TRH from samples clearly revealed that the proposed sensor can be applied into clinical analysis, quality control and a routine determination of drugs in pharmaceutical formulations.

Abstract: We conclude that significantly more patients in novel combination drug group compared to tramadol/paracetamol only group had a superior analgesic effect with lesser adverse reactions.
