Nosocomial dissemination of VIM-2-producing ST235 Pseudomonas aeruginosa in Lithuania

Abstract: Pseudomonas aeruginosa multidrug resistance, and particularly the production of carbapenemases linked to international high-risk clones, is of growing concern. While high levels of carbapenem resistance (>60 %) have been reported in Lithuania, so far, there is no information on the underlying mechanisms. Thus, the aim of this work was to determine the molecular epidemiology and prevalence of acquired carbapenemases among 73 carbapenem-resistant P. aeruginosa isolates recovered in a hospital from Kaunas, Lithua… Show more

“…Characterization of resistance mechanisms. AmpC hyperproduction, OprD deficiency, and the presence of horizontally acquired ␤-lactamases were first explored through previously established phenotypic and molecular (PCR) methods (26,69). The levels of expression, in comparison with the P. aeruginosa reference strain PAO1, of the genes encoding the chromosomal ␤-lactamase AmpC (ampC) and four P. aeruginosa efflux pump components (mexB [MexAB-OprM], mexD [MexCD-OprJ], mexF [MexEF-OprN], and mexY [MexXY-OprM]), were determined in selected isolates by real-time reverse transcription-PCR (RT-PCR) with an Eco real-time PCR system (Illumina), according to previously described protocols (70).…”

“…GES-5 producing P . aeruginosa strains have been isolated from patients in Brazil [ 30 ], Japan [ 14 ], Lithuania [ 31 ], South Africa [ 32 ], Spain [ 33 ] and Turkey [ 34 ]. Thus, it is important to monitor for GES-5 producing P .…”

“…The nine ST1047 P. aeruginosa strains showed highly homologous macrorestriction analysis patterns with 90.7% Dice similarity and they differed significantly from the one ST1203 strain (71.6% similarity). ST1047 was first reported as a VIM‐producing isolate associated with import from India in Norway ( P. aeruginosa PubMLST id: 746) and it has been reported in Europe and Asia . ST1203 was reported as a GES‐ and OXA‐producing isolate from a patient with an eye infection in India .…”
