Marine sources contain several bioactive compounds with high therapeutic potential, such as remarkable antioxidant activity that can reduce oxidative stress related to the pathogenesis of neurodegenerative diseases. Indeed, there has been a growing interest in these natural sources, especially those resulting from the processing of marine organisms (i.e., marine bio-waste), to obtain natural antioxidants as an alternative to synthetic antioxidants in a sustainable approach to promote circularity by recovering and creating value from these bio-wastes. However, despite their expected potential to prevent, delay, or treat neurodegenerative diseases, antioxidant compounds may have difficulty reaching the brain due to the need to cross the blood–brain barrier (BBB). In this regard, alternative delivery systems administered by different routes have been proposed, including intranasal administration of lipid nanoparticles, such as solid lipid nanoparticles (SLN) and nanostructured lipid carriers (NLC), which have shown promising results. Intranasal administration shows several advantages, including the fact that molecules do not need to cross the BBB to reach the central nervous system (CNS), as they can be transported directly from the nasal cavity to the brain (i.e., nose-to-brain transport). The benefits of using SLN and NLC for intranasal delivery of natural bioactive compounds for the treatment of neurodegenerative diseases have shown relevant outcomes through in vitro and in vivo studies. Noteworthy, for bioactive compounds obtained from marine bio-waste, few studies have been reported, showing the open potential of this research area. This review updates the state of the art of using SLN and NLC to transport bioactive compounds from different sources, in particular, those obtained from marine bio-waste, and their potential application in the treatment of neurodegenerative diseases.

Nanotechnology represents a magic wand to solve most problems related to improving drug effi ciency, one of the most important of these problems is the drug’s permeability through biological membranes. Rizatriptan benzoate was used for the treatment of acute migraine, it has poor permeability and is diffi cult to administer through the nose. This study aims to design a nanostructured lipid carrier containing rizatriptan benzoate as a trial to enhance its biological permeability. A high shear homogenization technique was utilized as a method of preparation; glyceryl monostearate and becs wax are used as solid lipids while oleic acid and castor oil were used as liquid lipids in diff erent ratios. Particle size analysis, polydispersity index, zeta potential, entrapment effi ciency, and loading effi ciency were considered the main criteria for the evaluation, meanwhile, the in-vitro release test was done for the formulas having smallest particle size. Moreover, the infrared spectroscopy and diff erential scanning calorimetry (DSC) are investigated for the selected formula. The obtained outcome revealed a signifi cant eff ect on the particle size and entrapment effi ciency upon enhancing the ratio of liquid lipid. Furthermore, changing the type of solid and liquid lipid leads to a dramatic alteration in the criteria of evaluation, also biphasic release pattern was seen. The infrared spectroscopy shows an intact rizatriptan benzoate, while DSC revealed a change of drug molecule to an amorphous state. In conclusion, a high shear homogenization can be used to formulate a successful nanostructured lipid carrier with good physical properties.

Intranasal delivery has great potential to cross the blood-brain barrier and deliver the drug molecule into the central nervous system faster than traditional methods. The olfactory neuronal and trigeminal pathways both are involved in intranasal delivery. The nano-technology is an innovative strategy for the nose to brain delivery. The mucoadhesive nanoemulsion formulation is a modified technology that increases the duration of drug accumulation and provides prolonged delivery at a targeted site. The nanoemulsion formulation oil, surfactant, and co-surfactant components maintain lower surface tension and particle coalescence. The globule dimension and zeta potential are affected in brain targeting. The globule size of the innovative formulation should be < 200 nm for drug permeation because, in humans, the average axon magnitude ranges from around 100 to 700. Furthermore, modified technology of nanoemulsion like nanogel and nanoemulsion in-situ gel provide a great advantage to cure neurodegenerative disorders. Therefore, focusing on the innovative pharmaceutical approaches of nanoemulsion in intranasal drug delivery, the current review provides insight into the applications of nanoemulsion in neurodegenerative disorders like Parkinson's disease, which are due to the depletion of dopamine in substania nigra resulting in cardinal motor activity bradykinesia and tremors. The review also touches upon the pathways for intranasal delivery of nanoemulsion, the pathogenesis of Parkinson's disease, and the future direction of the research on intranasal nanoemulsion.
