Noradrenaline Uptake Properties of the Anococcygeus Muscle of the Rat

Abstract: Noradrenaline (NA) uptake was measured in the anococcygeus by two methods: quantitative fluorescence microscopy and radioactive tracer techniques. A pharmacological analysis of the contribution of adrenergic nerve, smooth muscle, and nonspecific binding to collagen and basement membrane was attempted by studying the effects of desmethylimipramine and phenoxybenzamine on uptake. The importance of metabolic degradation in the process of NA accumulation was estimated by examining the effect of the combined use of… Show more

“…The accumulation of [3H]-noradrenaline with time in the rat anococcygeus muscle (Figure 4) shows a slower time course than that reported by Nash, Gillespie & Robertson (1974). However, they stretched the muscle into a flat sheet exposing the maximum surface to the medium, whereas in the present experiments the muscles were free floating and were contracted by the noradrenaline (10-6 M).…”

“…With these low concentrations uptake is likely to be mainly into adrenergic nerves, with no accumulation within smooth muscle (Nash et al, 1974). The results obtained in these experiments are illustrated in Figure 1 where maximum tissue:medium ratios of 5.4 ± 0.8 (s.e.…”

“…The accumulation of noradrenaline from low concentrations of the amine by the rat anococcygeus muscle is predominantly neuronal (Nash, Gillespie & Robertson, 1974). Furthermore, following incubation with low concentrations of tritiated noradrenaline, all the radioactivity in the rat anococcygeus muscle behaves as authentic noradrenaline (Doggrell & Woodruff, 1977).…”
