Noncovalent Liposome Linkage and Miniaturization of Capsosomes for Drug Delivery

Abstract: We report the synthesis of poly(methacrylic acid)-co-(oleyl methacrylate) with three different amounts of oleyl methacrylate and compare the ability of these polymers with that of poly(methacrylic acid)-co-(cholesteryl methacrylate) (PMA(c)) to noncovalently anchor liposomes to polymer layers. We subsequently assembled ∼1 μm diameter PMA(c)-based capsosomes, polymer hydrogel capsules that contain up to ∼2000 liposomal subcompartments, and investigate the potential of these carriers to deliver water-insoluble d… Show more

“…Thiocoraline (TC) loaded liposomes ( TC L) were assembled by adding 100 lL (0.1 mg/mL) TC in chloroform to the lipid mixture prior to drying. This TC concentration was chosen to ensure the highest amount of incorporated cargo into the lipid membrane of the liposomes based on our previous reports [29,30]. Thus, increasing TC concentration above 0.1 mg/mL does not results in an increase in the amount of liposome-loaded TC.…”

“…Further, we have demonstrated the functionality of capsosomes by performing two different triggered enzymatic assays14, 17 and a two‐step enzymatic catalytic reaction 18. The absence of inherent cytotoxicity of capsosomes (by performing a cell viability assay)19 makes capsosomes promising candidates for biomedical applications.…”
