Noncoding Effects of Circular RNA CCDC66 Promote Colon Cancer Growth and Metastasis

Abstract: Circular RNA (circRNA) is a class of non-coding RNA whose functions remain mostly unknown. Recent studies indicate circRNA may be involved in disease pathogenesis, but direct evidence is scarce. Here we characterize the functional role of a novel circRNA, circCCDC66, in colorectal cancer (CRC). RNA-Seq data from matched normal and tumor colon tissue samples identified numerous circRNAs specifically elevated in cancer cells, several of which were verified by quantitative RT-PCR. CircCCDC66 expression was elevat… Show more

“…As reported, circRNAs could function as a miRNA sponge to regulate the gene expression [26, 27] and be involved in cancer processes [28-30]. To dissect the selected circRNAs’ potential functions, the circRNA and miRNA interactions were predicted by using Arraystar’s homemade miRNA target prediction software based on TargetScan and miRanda.…”

“…[35] circRNACCDC66 was shown to accelerate the proliferation and metastasis of colon cancer, [36] circLARP4 regulated LARP4 expression by competitively binding to miR-424-5p in gastric cancer, [37] and circLMO7 was involved in the regulation of myogenic differentiation in cattle. circRNAs have been shown to play important roles in various cancers.…”

“…In the study field of tumorigenesis and progression, recent studies have revealed that certain circRNAs are deregulated in many kinds of human cancers through high‐throughput RNA sequencing, such as gastric cancer, breast cancer, basal cell carcinoma, and cutaneous squamous cell carcinoma . Furthermore, several aberrant circRNAs have been identified as oncogenes or tumor suppressors . However, the expression and functions of most deregulated circRNAs remain largely unknown, and the mechanisms are rarely uncovered.…”
