Noncoding dsRNA induces retinoic acid synthesis to stimulate hair follicle regeneration via TLR3

Abstract: How developmental programs reactivate in regeneration is a fundamental question in biology. We addressed this question through the study of Wound Induced Hair follicle Neogenesis (WIHN), an adult organogenesis model where stem cells regenerate de novo hair follicles following deep wounding. The exact mechanism is uncertain. Here we show that self-noncoding dsRNA activates the anti-viral receptor toll like receptor 3 (TLR3) to induce intrinsic retinoic acid (RA) synthesis in a pattern that predicts new hair fol… Show more

“…In the latter study, addition of a single local dose of dsRNA, 3 days after skin wounding in wild-type mice, increased the number of regenerated follicles, while addition of RNAse III to the site reduced the number of follicles [20]. TLR3 activation by dsRNA induced aldehyde dehydrogenase 1 family member A3 (ALDH1A3), an enzyme which converts retinol to retinoic acid in keratinocytes, and accumulation of RA was required for WIHN [70].…”

“…The samples were prepared from arm skins before (as controls) and 7 days after the treatment with a 1550 nm non-ablative fractional CO2 laser (as treated). The original data were published by Kim et al as "Noncoding dsRNA induces retinoic acid synthesis to stimulate hair follicle regeneration via TLR3" 13 in Nature Communications. We determined and discussed the role of the central nodes of the protein-protein interaction network of the DEGs to highlight "cytokine-mediated signaling pathway" as a major affected individual by laser irradiation.…”

“…The microarray results of a document entitled "Noncoding dsRNA induces retinoic acid synthesis to stimulate hair follicle regeneration via TLR3" 12 were downloaded from Gene Expression Omnibus (GEO). The Data are presented as GSE131789/GPL15207 for 16 women that are treated to improve photo aging.…”
