Non-invasive Brain Delivery and Efficacy of BDNF in APP/PS1 Transgenic Mice

Abstract: Neurotrophic factors such as brain-derived neurotrophic factor (BDNF) and nerve growth factor (NGF) have been demonstrated for their potential as a neuroregenerative treatment of Alzheimer's disease (AD). Unfortunately, most proteins cannot be effectively delivered into the brain from the blood stream due to the presence of the blood-brain barrier (BBB). In this study, we delivered BDNF using ADTC5 as BBB modulator (BBBM) into the brains of transgenic APP/PS1 mice, a mouse model for AD. As controls, two groups… Show more

“…An in vitro study shows that Aβ plays a role in reducing BDNF level (Zheng et al, 2010), which may lead to impaired synaptic plasticity. Intravenous infusion of BDNF with ADTC5, a blood brain barrier modulator, increased the level of BDNF in the hippocampus of APP/PS1 mice and increased BDNF was associated with higher efficacy in NOR and Y-maze tasks (Kopec et al, 2020). Neonatal TS from day 8-14 has proven to be beneficial in increasing BDNF level, and TS from day 1-7 and day 15-21 hasve been shown to reduce glucocorticoid receptors in the hippocampus in adult rats (Antoniazzi et al, 2016).…”

“…However, there might be an alternative pathway to ensure sufficient remyelination or decrease axonal loss without upregulation of ERK. Increased expression of BDNF and its corresponding receptor TrkB could possibly enhance remyelination [61].…”

“…Therefore, some BBB modulators, such as cadherin peptides (ADTC5), have been used to improve BDNF’s efficiency in crossing the BBB. Intravenous injection of BDNF with ADTC5 in transgenic APP.PS1 mice improved the cognitive performance of these AD mice compared to BDNF alone [ 103 ].…”
