Nog2p, a putative GTPase associated with pre-60S subunits and required for late 60S maturation steps

Abstract: Eukaryotic ribosome maturation depends on a set of well ordered processing steps. Here we describe the functional characterization of yeast Nog2p (Ynr053cp), a highly conserved nuclear protein. Nog2p contains a putative GTP-binding site, which is essential in vivo. Kinetic and steady-state measurements of the levels of pre-rRNAs in Nog2p-depleted cells showed a defect in 5.8S and 25S maturation and a concomitant increase in the levels of both 27SB S and 7S S precursors. We found Nog2p physically associated wit… Show more

“…Disruption of Nog1 function through RNA interference led to a dramatic decrease in the levels of free 60 S particles and the appearance of an atypical rRNA intermediate in which ITS2 was not cleaved (40). Likewise, depletion of Nug2 resulted in a dramatic decrease in 5.8 S rRNA levels and accumulation of the 7 S precursor as well as nucleolar accumulation of ITS2-containing precursors (6). These GTPases may act in concert with the Rea1 ATPase to coordinate the structural reorganization of late pre-60 S ribosomes, prior to the irreversible step of nuclear export.…”

“…In the last few years, the maturation of 40 S and 60 S pre-ribosomes has been extensively analyzed by purification of pre-ribosomal particles (5)(6)(7)(8)(9)(10). Interestingly, a large number of non-ribosomal proteins were identified in pre-60 S particles without an assigned function in RNA metabolism.…”

“…Yeast GTPases Nug1p and Nog2p/Nug2p are required for the maturation and export of the large ribosomal subunit (Bassler et al, 2001;Saveanu et al, 2001). To test whether Drosophila NS1 is required for large ribosomal subunit maturation/transport, we first examined transgenic larvae that expressed GFP-tagged RpL11 (GFP-RpL11/ϩ; da-GAL4/ϩ).…”

“…A favored approach has been to purify these complexes from cell lysates [1][2][3][4] . But the limitations of presently available purification techniques pose a major hurdle to the study of RNA processing and export.…”
