NMR-Based Metabonomic Investigation of Heat Stress in Myotubes Reveals a Time-Dependent Change in the Metabolites

Abstract: NMR-based metabonomics was applied to elucidate the time-dependent stress responses in mouse myotubes after heat exposure of either 42 or 45 degrees C for 1 h. Principal component analysis (PCA) revealed that the gradual time-dependent changes in metabolites contributing to the clustering and separation of the control samples from the different time points after heat stress primarily are in the metabolites glucose, leucine, lysine, phenylalanine, creatine, glutamine, and acetate. In addition, PC scores reveale… Show more

“…Technological developments are the driving force behind advances in scientific knowledge. Recent advances in the two analytical platforms of mass spectrometry (MS) and nuclear magnetic resonance (NMR) spectroscopy have driven forward the discipline of metabolomics [28][29][30][31][32]. Technological advances in NMR and mass spectrometry have opened a new chapter in biochemistry by using metabolomics as an approach to study metabolism and its regulation in relation to genetic, disease, and environmental factors.…”

“…This is because metabonomics systemically detects the holistic and dynamic metabolic responses of an integrated biological system to the changes of both endogenous and exogenous factors . Such an approach has been already successfully applied in understanding the metabolic responses to stress factors and systems metabolic reprogramming underlying the tolerance and acclimation to a harsh environment. It is particularly interesting to note that the metabonomics approach has also been proved powerful in analyzing the chemical compositions of processed products of P. trituberculatus …”

“…Metabonomic information is obtained by maximum data capture from samples using Nuclear Magnetic Resonance (NMR), Mass Spectrometry (MS), High-Performance Liquid Chromatography (HPLC) and Gas Chromatography (GC) followed by multivariate data analysis. 1 H-NMR-based metabonomics have been suggested as an attractive tool for the investigation of the effects of depression [17], [18] due to its non-destructive nature and rapid collection of data.…”
