Nitric Oxide Involvement in Cardiovascular Dysfunctions of Parkinson Disease

Abstract: Parkinson’s disease (PD) is characterized by the loss of dopaminergic neurons in the substantia nigra, causing motor changes. In addition to motor symptoms, non-motor dysfunctions such as psychological, sensory and autonomic disorders are recorded. Manifestations related to the autonomic nervous system include the cardiovascular system, as postural hypotension, postprandial hypotension, and low blood pressure. One of the mediators involved is the nitric oxide (NO). In addition to the known roles such as vasodi… Show more

“…Nitric oxide synthase (NOS) is the enzyme responsible for NO synthesis. Excessive NO production is associated with a wide range of pathophysiological states including multiple sclerosis, 4 sepsis, 5 Parkinson's disease, 6 metabolic dysregulation, 7 and cancer. 8 The use of NOS inhibitors may directly inhibit NO production, however, simultaneously blocking its beneficial effects.…”

“…NO can lead to the formation of peroxynitrite (ONOO − ), which produces the interaction of NO with superoxide radicals [ 31 , 32 ]. NO can also inhibit the mitochondrial respiratory chain, especially in complex IV [ 33 ].…”
