Nitrate reductase activity in nodules of pea inoculated with hydrogenase positive and hydrogenase negative strains of Rhizobium leguminosarum

Abstract: The nitrate reductase (NR, EC 1.6.6.1) activity in root nodules formed by hydrogenase positive (Hup+) and hydrogenase negative (Hup−) Rhizobium leguminosarum strains was examined in symbioses with the pea cultivar Alaska (Pisum sativum L.), Rates of activity were determined by the in vivo assay in nodules from plants that were only N2‐dependent or grown in the presence of 2 mM KNO3. The rates varied widely among strains, regardless of the Hup phenotype of the R. leguminosarum strain used for inoculation, but t… Show more

“…The strong influenee (80% effect) of the plant genotype upon the manifestation of NRA in roots and nodules and also in shoots (data not shown) of Vicia faba supports previous reports on inter-and intraspecific variability in nitrogen use between legumes (Becana and Sprent 1987, Ligero et al 1987b, Peoples et al 1987.…”

“…Nodule and root NRA was measured both in vivo and in vitro. The in vivo method was based on that described by Crafts-Brandner and Harper (1982) and Ligero et al (1987a). Subsamples of 0.25 g (whole nodules) and 0.5 g (2 to 3 mm root pieces) were placed in vials containing 5 ml of ice-cold incubation medium consisting of 0.1 M K-phosphate buffer (pH 7.0), 50 mM KNO3, 1 mM Na^EDTA, 2% (v/v) M-propanol and 0.01% (v/v) Triton X-100 as wetting agent.…”

“…Recent reports indicate that free-living rhizobia can reduce NOj simultaneously to NHJ (assimilation) and to N, (denitrifieation) (Vairinhos et al 1986). Ligero et al (1987) have observed that pea nodules with bacteroids expressing uptake hydrogenase activity (phenotypes Hup*) show lower NRAs in vivo than nodules containing Hup" bacteroids. They suggest that the presence of higher NRAs can counteract the lack of an hydrogenase system.…”

“…Nitrate reductase (NR) activity was determined in leaves by the modified in vivo method (Jaworski, 1971;Ligero et al, 1987;Karwat et al, 2019). The NR activity was determined in the flag leaf, which was harvested between Z51 and Z53 stages (Zadoks et al, 1974).…”
