NIR triggered PLGA coated Au-TiO2 core loaded CPT-11 nanoparticles for human papillary thyroid carcinoma therapy

Abstract: MDR (multi-drug resistance) is one of the significant deterrents of effective chemotherapy for malignant growth. One of the powerful ways to deal with defeat of the MDR is to utilize inorganic nanoparticle-intervened tranquilize conveyance to build the medication aggregations in cancerous growth cells. In this work, we have developed the presentation that is accurately made of medication conveyance framework dependent on the TiO 2 nanoparticles stacked CPT-11 to defeat the thyroid malign… Show more

“…Briefly, BALB/c nude mice bearing KTC-1 tumors (≈150 mm 3 ) were divided into two groups (n = 3) and intravenously injected with Cy5-labeled PPR NC and free siNC-Cy5, respectively. At different time intervals (6,12,24,36, and 42 h) after injection, fluorescence images of mice were taken using the IVIS SPECTRUM (PerkinElmer) in vivo imaging system. The heart, liver, spleen, lungs, kidneys, and tumor were collected and photographed as mentioned above at 36 h after injection.…”

“…Ultrasound (US)-guided radiofrequency ablation has been recruited as an alternative therapy for PTC instead of surgery in recent years, which promotes the life quality of patients to some extent. However, with the rise of nanomedicine, photothermal therapy (PTT) has emerged, which is based on ablation of tumor tissues by photothermal conversion nanomaterials upon near-infrared (NIR) laser irradiation with minimal invasiveness and accuracy. In addition, photothermal nanomaterials are also excellent photoacoustic imaging (PAI) agents that are well suited for the detection of in vivo nanoparticle distribution and guidance of laser irradiation, providing a more safe and promising method for PTC treatment. However, lymphogenic metastasis and recurrence along with PTC tumor ablation is still an urgent issue to be addressed.…”

“…In terms of amplifying the immune reactivity to alleviate tumor immune escape, nanoplatforms are often coated with polymeric materials aiming at different targets. In this context, various synthetic polymers, such as polystyrene [ 51 ], poly(lactic- co -glycolic acid) (PLGA) [ 52 , 53 ], and poly(beta-amino esters) (PBAEs) [ 54 ], in the order of non-, slowly-, and rapidly-degradable, present intrinsic immunostimulatory properties via activating dendritic cells (DCs) and causing an increase of inflammatory cytokines (IL-1β) that supports early immune responses [ 55 ]. Compared to polystyrene and PBAEs, PLGA-based nanoparticles are more frequently used to serve not only as carriers but also as agents that elicit or modulate the immune response.…”

“…The American Thyroid Association guidelines recommend docetaxel/paclitaxel and platinum in ATC, despite yielding no benefit to survival in advanced ATC [ 78 ]. One of the key problems preventing the successful treatment of malignant tumors is multidrug resistance (MDR) [ 79 ]. The construction of nanoparticles for precise targeted delivery of chemotherapeutic drugs is expected to solve this problem, and the construction of optical diagnostic imaging, phototherapy, and chemotherapeutic drug delivery in a multimodal nanoplatform has become a popular topic for research.…”
