New trends in the treatment of bone metastasis

Abstract: Bone metastasis is often the penultimate harbinger of death for many cancer patients. Bone metastases are often associated with fractures and severe pain resulting in decreased quality of life. Accordingly, effective therapies to inhibit the development or progression of bone metastases will have important clinical benefits. To achieve this goal understanding the mechanisms through which bone metastases develop and progress may provide targets to inhibit the metastases. In the past few years, there have been a… Show more

“…However, development efforts have increasingly included bone metastases [ 18 ]. Bisphosphonates act by blocking the RANK-RANK ligand interaction [ 19 ]. Second-generation or aminobisphosphonates (ie, zoledronate) may also be useful due to potent RANK inhibition and indirect inhibition of the Ras signaling pathway.…”

“…Bone metastasis causes consequences, such as bone pain. Bone fractures and nerve compression, seriously affect the patient's quality of life (18,19). Thus, it is urgent to develop accurate predictors and illuminate the mechanisms of PCa distant metastasis.…”

“…Once the tumor has developed bone metastases, the disease in most cases is incurable. However, surgical resection of these metastases and stabilization of pathologic fractures are generally considered a valuable palliative treatment [8]. The adjuvant chemotherapy is effective to slow down tumor growth and metastasis outgrowth.…”
