New Treatment Options Using 5-HT3 Receptor Antagonists in Rheumatic Diseases

Abstract: In vitro studies have shown that a blockade of 5-HT3 receptors brings about a reduction of tumor necrosis factor, IL-1 beta, IL-2, IL-6 as well as a decrease in prostaglandins. Clinical trials have provided evidence of pain reduction in a subgroup of fibromyalgia syndrome and, moreover, have demonstrated that tropisetron injected locally for insertion tendinoses and myofascial syndromes with associated trigger points leads to an alleviation of pain that is comparable to injections with the combination of corti… Show more

“…48,49 Laboratory studies have also shown the inhibition of serotonin receptors which is associated with the reduction of TNFα, IL1β, IL6, IL2, and prostaglandins. 37 In addition Ginger have thermogenic properties and increases energy expenditure by enhancing the thermic effect of food.…”

“…After development of other indications for 5HT 3 receptor antagonists, recently, immunomodulatory and anti-inflammatory properties have been observed for these antagonists [13,14]. It was found that tropisetron inhibits TNFα and IL-1β release in monocytes, PGE 2 release from synovial cells and also diminishes calcineurin-induced T-cell activation [31][32][33] and modulates Th1 cytokines [34].…”

“…The efficacy of this drug has also been attributed to its antiphlogistic effect, which may be related to the inhibited release of substance P and other neuropeptides from nociceptors, and the blocked release of phlogistic substances from macrophages, monocytes, etc 22,69,70,91 .…”
