New 99mTc(CO)3 Mannosylated Dextran Bearing S-Derivatized Cysteine Chelator for Sentinel Lymph Node Detection

Abstract: The aim of the present study is to synthesize new mannosylated dextran derivative that can be labeled with Tc-99m for potential use in sentinel lymph node detection (SLND). The compound was designed to have a dextran with molecular weight of 10 kDa as a backbone, mannose for binding to mannose receptors of the lymph node and S-derivatized cysteine as a suitable chelator for labeling with [(99m)Tc(H(2)O)(3)(CO)(3)](+) precursor. Reaction of allyl bromide with dextran (MW 11800) yielded the intermediate allyl-de… Show more

“…5 Application of BASPs opens unique opportunities for nuclear medicine: on the one hand, BASPs show their own immunostimulatory activity, and on the other hand, they allow preparation of the complex conjugates with grafted vectors (low MW biologically active substances providing target receptor binding to the tumor cells). 6,7 The use of such biologically active substances labelled with emitters of the Auger and conversion electrons (ie, radionuclides, often used in single photon emission computer tomography or PET, such as 44m Sc, 67 Ga, 68 Ga, 99m Tc, 111 In, etc) allows development of novel target radiopharmaceuticals suitable not only for early stage diagnostics but also for performing effective radiotherapy.…”

“…Attempts to isolate pure imidate by flash chromatography were unsuccessful due to decomposition, although the more stable 39 was isolable. Literature reports of the coupling of imidate 40 to amines utilize the crude reaction mixture described above and require a five‐fold excess of imidate to amine, making the synthesis of 1 rather inefficient. The characterization of 1 is also lacking with the only data reported being limited 1 H NMR data at 200 MHz .…”

“…It is well known that S ‐alkylated cysteine derivatives bind efficiently with the tricarbonyl core via its NSO donor‐atom set to provide stable and neutral complexes. Coordination affords two five‐membered and one six‐membered ring providing thermodynamically stable hexacoordinated complexes . The proposed structure for the final complex is shown in Figure .…”
