New Sesquiterpene Lactones of Eremophilane-Type from Ligularia Fauriei (Fr.) Koidz.

Abstract: Two new sesquiterpene lactones of eremophilane-type were isolated from Ligularia Fauriei (Fr.) Koidz., and their structures including absolute stereochemistry were shown to be 8β-hydroxyeremophil-7(11)-ene-12,8α;14β,6α-diolide (1) and eremophil-7(11)-ene-12,8α;14β,6α-diolide (2), respectively. The isolation of the two known lactones, 6β,8β-dihydroxyeremophil-7(11)-en-12,8α-olide (3) and 6β-hydroxyeremophil-7(11)-en-12,8α-olide (4), is also described. The photosensitized autoxidation of the known furanoeremophi… Show more

“…
In our ongoing investigation into bioactive compounds from the genus Ligularia (Compositae) plants [1][2][3][4][5][6][7] we have studied the roots of Ligularia tongolensis (FRANCH) HAND-MAZZ collected in mountainous areas (altitude: 3600 m) in southwestern China. Ligularia tongolensis is used as a traditional folk medicine in China and its roots can reduce phlegm, relieve coughing and cure pulmonary tuberculosis, urinary track blockages, common cold, and pharyngitis.

However, the chemical constituents of this plant have not been reported up until now.

“…4 As for relative configurations, CH 3 -14, CH 3 -14¢, CH 3 -15 and CH 3 -15¢ were all assigned as b orientation on the basis of biogenetic consideration of eremophilane derivatives from Ligularia species. 10 The observed homoallylic coupling between CH 3 -13 (d, J = 0.9 Hz) with H-6a (brd, J = 13.0), and CH 3 -13¢ (brs) with H-6¢a (brd, J = 12.4) suggested the 12(8b)-olides configurations of two substructures. 11 CH 3 -14), 0.88 (d, J = 6.9, CH 3 -15¢) and 0.76 (s, CH 3 -14¢) further confirmed the 8b configuration of two lactone groups.…”

“…As for stereochemistry in 1, the H 3 -14 and H 3 -15 were assigned to be β configuration in biogenetic consideration. 14 The NOESY cross peaks observed between H3-14 and H-10 ( Figure 2) implied a cis-junction for the two rings. 9 The methoxy group at C-8 was deduced to be β orientation based on the downfield shifts of H3-14 compared to H3-15.…”
