New polyene macrolide family produced by submerged culture of Streptomyces durmitorensis

Abstract: A new polyene macrolide family, closely related to the pentaene macrolide antibiotic roflamycoin, was isolated from the both fermentation broth and biomass of Streptomyces durmitorensis wild-type strain MS405. The main compound was identified by NMR and Fourier transform ion cyclotron resonance mass spectrometry as 32,33-didehydroroflamycoin (1; DDHR). Additional four structurally related compounds were determined solely by MS analysis. DDHR induces cell death by apoptosis in various cancer cell lines as demon… Show more

“…1b). The absorption spectra were in good correlation with previously reported maxima at 260 nm, 363 nm and a shoulder at 375 nm (Stodulkova et al 2011). Upon purification using flash chromatography, polyene-containing fraction was analysed by liquid chromatography followed by tandem mass spectroscopy (LC-MS), and it was confirmed that major product of the polyene family was DDHR ( Fig.…”

“…Stodulkova et al reported that DDHR colocalized with late endosomal/lysosomal markers in HeLa cells and induced apoptosis after 4 h of treatment with IC50 (concentration of the drug causing 50% inhibition of the cell viability) varying between 60 and 100 lmol l À1 (Stodulkova et al 2011). The cytotoxicity assay on fibroblast cell line presented here confirmed the cytotoxic effect of DDHR at relatively high concentrations (Fig.…”

“…rapamycin (a triene), nystatins (tetraene), filipins (pentaene) and amphotericin B (a heptaene) (Eva Stodůlková et al . ). A pentaene macrolide 32, 33–didehydroroflamycoin (DDHR) (also known as Dhanyabadomycin), which is a new derivative of Roflamycoin (Synonym: flavomycoin) has been reported by Eva Stodulkova et al .…”

“…To date, more than 200 known polyene macrolide antibiotics (e.g., rapamycin, nystatins, filipins and amphotericin B) have been isolated and characterized, most of which are produced by the genus Streptomyces [4]. In the course of our screening program for new microbial natural products, we isolated a new polyene macrolide antibiotic antifungalmycin 702 from S .…”
