“…The determination of the percentage of viable cells after the various treatments with the F-PSEO and D-PSEO was performed by using the dual DNA intercalating fluorescent dyes kit from EMD Millipore Bioscience (Muse TM Cell Count & Viability Assay) as previously described in our reports. [28][31] Briefly, 3 9 10 5 cells/ml of the leukemic (HL-60, Jurkat, and K562) in RPMI-1640 medium, or 4 9 10 4 cells/ml of the solid tumor cells (MCF-7, HepG2, HT-1080, and Caco-2) in DMEM medium, were seeded in each well of a 48 well-plate and incubated for 24 h. Subsequently, the cultured cells were treated with increasing concentrations of F-PSEO or D-PSEO (0.0 -10.0 lg) and incubated for additional 24 h in a 5% CO 2 -cell culture incubator. At the end of the exposure period, both the suspension and adherent (after trypsinization) cells were transferred to Eppendorf tubes and washed twice with a 1 ml of PBS.…”
