“…The determination of the percentage of viable cells after the various treatments with the F-PSEO and D-PSEO was performed by using the dual DNA intercalating fluorescent dyes kit from EMD Millipore Bioscience (Muse TM Cell Count & Viability Assay) as previously described in our reports. This study was performed to determine the chemical composition, antioxidant and cytotoxic effects of essential oils extracted from the aerial parts of fresh (F-PSEO) and air-dried (D-PSEO) Pallenis spinosa. The composition of the oils was analyzed by gas chromatography (GC) and GC/mass spectrometry, the antioxidant activity by free radical scavenging and metal chelating assays, and their cytotoxicity by a flow cytometry analysis. The primary components in both oils were sesquiterpene hydrocarbons and oxygentated sesquiterpenes. F-PSEO contained 36 different compounds; α-cadinol (16.48%), germacra-1(10),5-diene-3,4-diol (14.45%), γ-cadinene (12.03%), and α-muurolol (9.89%) were the principal components. D-PSEO contained 53 molecules; α-cadinol (19.26%), δ-cadinene (13.93%), α-muurolol (12.88%), and germacra-1(10),5-diene-3,4-diol (8.41%) constituted the highest percentages. Although both oils exhibited a weak radical scavenging and chelating activity, compared to α-tocopherol and ascorbic acid, D-PSEO showed a 2-fold greater antioxidant activity than F-PSEO. Furthermore, low doses of F-PSEO were able to inhibit the growth of leukemic (HL-60, K562, and Jurkat) and solid tumor cells (MCF-7, HepG2, HT-1080, and Caco-2) with an IC range of 0.25 - 0.66 μg/ml and 0.50 - 2.35 μg/ml, respectively. F-PSEO showed a ca. 2 - 3-fold stronger cytotoxicity against the tested cells than D-PSEO. The potent growth inhibitory effect of the plant essential oil encourages further studies to characterize the molecular mechanisms of its cytotoxicity.

Under natural conditions, the plants are, usually, surrounded by a series of potential enemies. They had created strategies of defense against pathogens and herbivores' attacks, allowing its perpetuation throughout evolution. These defense mechanisms are closely associated with the synthesis of secondary metabolites, which are also worldwide used in several areas of industry. This bibliographic review was conducted aiming to better understand how plants synthesize these substances in response to biotic or abiotic stimuli. The results discussed here revealed that synthesis of plant secondary metabolites is dependent on changes in several metabolic pathways, being often directly associated with the primary metabolism. Injury by herbivores or pathogens, temperature, CO 2 levels, solar radiation and drought, are the factors related to the environment that express the most significant signs of inhibition or synthesis of plant secondary metabolites. Global climatic changes recently observed can affect the metabolic pathways network and, consequently, secondary metabolites synthesis. Nowadays, genomic tools have been useful alternatives that are leading to a new revolution of plant breeding, allowing the overexpression or inhibition of these substances. Some limitations and challenges to be achieved upon the dynamics of secondary metabolite synthesis in plants are presented.

Objective Experimental studies reported that some plants in the genus of Moraea (Iridaceae family) show anticancer multiform and HepG2 liver cancer cells. Methods The cells were incubated potential. This study aimed to evaluate the effects of Moraea sisyrinchium on U87 Glioblastoma for 24 with hydro alcoholic extract of the stem, flower, and bulb of Moraea sisyrinchium. The cell proliferation, cell cycle, and migration were determined by MTT assay, propidium iodide staining, and scratch assay, respectively. Oxidative stress was evaluated by determining the levels of reactive oxygen species and lipid peroxidation. The activity of matrix metalloproteinases (MMPs), the expression of Bax and Bcl-2 proteins, and angiogenesis were evaluated by the gelatin zymography, Western blotting, and model of chick embryo chorioallantoic membrane, respectively. Results The extracts of the flower, stem, and bulb significantly decreased the proliferation of HepG2 and U87 cells. This effect was more for U87 than for HepG2 and for bulb and stem than for flower. In U87 cells, the bulb extract increased oxidative stress, cell cycle arrest, and the Bax/Bcl-2 ratio. Also, this extract suppressed the migration ability of HepG2 and U87 cells, which was associated with the inhibition of MMP2 activity. In addition, it significantly reduced the number and diameter of vessels in the chorioallantoic membrane. Liquid chromatography-mass spectrometry revealed the presence of xanthones (bellidifolin, mangiferin), flavonoids (quercetin, luteolin), isoflavones (iridin,tectorigenin),and phytosterols (e.g., stigmasterol) in the bulb extract. Conclusion The extract of Moraea sisyrinchium bulb decreased the proliferation and survival of cancer cells by inducing oxidative stress. The extract also reduced the migration ability of the cells and inhibited angiogenesis.
