New Insights into Peptide–Silver Nanoparticle Interaction: Deciphering the Role of Cysteine and Lysine in the Peptide Sequence

Abstract: We studied the interaction of four new pentapeptides with spherical silver nanoparticles. Our findings indicate that the combination of the thiol in Cys and amines in Lys/Arg residues is critical to providing stable protection for the silver surface. Molecular simulation reveals the atomic scale interactions that underlie the observed stabilizing effect of these peptides, while yielding qualitative agreement with experiment for ranking the affinity of the four pentapeptides for the silver surface.

“…Like AuNPs, AgNPs are small in size, have high surface area-to-volume ratios, and display unique electrical and optical properties resulting in high reactivity and chemical stability [23]. In addition, thiol chemistry can be utilized for direct conjugation to the AgNP, which allows for an easy method of conjugation that has been shown to increase both the stability and activity of the bound AMP [114][115][116]. However, AgNPs generally exhibit high cytotoxicity and are thus not necessarily a great delivery vehicle for improving AMP tolerability.…”

“…To improve their efficacy and stability in biomedical and other applications, the nanoparticles often need to be coated with non-toxic and non-inflammatory capping agents like collagen, peptides, and biopolymers, preferably without significantly changing their size, shape, and antimicrobial properties [ 3 , 15 , 16 , 17 , 18 , 19 ]. Some findings concerning the action of AgNPs suggest that the bactericidal mode can be influenced by the stability of the particles [ 5 ], and as one example AgNPs conjugated with poly-lysine were less effective when compared to collagen-capped AgNPs due to aggregation.…”

“…Though the exact mechanism of AgNP binding to Msr1 receptor is not known, we speculate that it might be unspecific binding to thiol (-SH) groups. AgNP have a strong affinity for cysteine tiol (-SH) groups of proteins [ 46 ] that are abundant in Msr1 scavenger receptor [ 47 ].…”
