New Dual Monoclonal ELISA for Measuring Plasma Osteopontin as a Biomarker Associated with Survival in Prostate Cancer: Clinical Validation and Comparison of Multiple ELISAs

Abstract: BACKGROUND:A previously developed monoclonal/ polyclonal ELISA (Mono/Poly) to detect plasma concentrations of osteopontin (OPN) was shown to provide prognostic information in breast, prostate, and other cancers. Here we describe the clinical validation of a new dual monoclonal (Dual Mono) assay. We compared both assays with 4 assays that recognize defined regions of OPN protein (dual polyclonal systems 5-1, 4-1, 4-3 and polyclonal-monoclonal system 1-3).

“…These modifications can be cell type-specific (Christensen et al 2007), may depend on other physiological and pathological factors and may impact both OPN structure and function (Anborgh et al 2009;Zhang et al 2007). This review will focus on OPN's role in cancer and how splice variants and posttranslational modifications may influence its effects in cancer cells.…”

“…OPN is found indifferent types of tissues and body secretions [1] with highest concentrations in milk [2]. OPN is a chief component in several of physiological processes such as inhibition of ectopic calcification, bone remodeling and immune modulatory functions [3,1,4], indicating that milk OPN could be an important factor in the development of the immune system of infants. Milk OPN is highly resistant to adverse conditions and heat treatment at 90 ⁰C does not bear any effect on the post-translational modifications of the protein [5].…”

“…It shows advantages of simplicity, high sensitivity, and easy standardization. However, ELISA method is not suitable for small molecule or trace substance detection [1][2][3][4][5]. In this article, the focus is on novel technologies and their potential for small molecule biomarkers, even for living cell analysis.…”
