New directions of miniaturization within the proteomics research area

Abstract: An overview of the current trends within protein expression profiling is given where multidimensional separation of both gel‐ and liquid‐phase techniques linked to mass spectrometry is viewed as a major route in the global proteome mapping. A clear trend in these biochemical developments is the effort to sequence and identify low‐abundant protein expressions where assay miniaturization and integrated sample processing plays a central role. Two areas of miniaturization within the proteomics field are addressed:… Show more

“…However, more easily automated LC techniques, often combined with MS, have begun to replace 2-D PAGE [26,27]. Gelfree proteomics can also more easily take advantage of labeling of the samples with stable isotopes [28] or chipbased techniques [29,30].…”

“…Recently, we presented alternatives of microchip configuration and geometries, and their respective performance in terms of operational stability, sensitivity and microfluidic characteristics [53][54][55][56][57][58]. In the new development, the sample loading and washing was separated and performed off-line from the actual extraction and sample spotting onto the MALDI target.…”

“…Different configurations such as single and triple quadrupole, TOF, IT, and ultrahigh-resolution Fouriertransform ion cyclotron resonance (FTICR) mass spectrometers were adapted to chip-based ESI and contributed significant benefits for various studies. However, as nicely illustrated already by a number of reviews [8,11,[20][21][22] the general areas of implementation and applications of chip technologies either silicon-based, glass, or polymer microchips have by far been primarily genomics, drug discovery, and proteomics.…”
