New chlorinated xanthone and anthraquinone produced by a mangrove-derived fungus Penicillium citrinum HL-5126

Abstract: Two new chlorinated metabolites 4-chloro-1-hydroxy-3-methoxy-6-methyl-8-methoxycarbonyl-xanthen-9-one (1) and 2'-acetoxy-7-chlorocitreorosein (2), together with three known compounds (3-5), were obtained from the EtOAc extract of the endophytic fungus Penicillium citrinum HL-5126 isolated from the mangrove Bruguiera sexangula var. rhynchopetala collected in the South China Sea. Their structures were elucidated by the detailed analysis of comprehensive spectroscopic data. All compounds were evaluated for their … Show more

“…However, we cannot exclude a possibility that this metabolite might be also produced by other members of the Pleosporales of which we identified a broad diversity using DNA sequencing . Nine metabolites (3‐nitropropionic acid, asperglaucide, chlorocitreorosein, citreorosein, emodin, linamarin, lotaustralin, physcion and tryptophol) could be derived from plants and fungi . Usnic acid and physcion are produced by lichen, a composite organism consisting of a symbiosis between a fungus and an algae or cyanobacterium.…”

“…For example, penicillixanthone isolated from a marine-derived fungus Aspergillus terreus from the gorgonian coral Echinogorgia aurantiaca exhibited potent antifouling activity against larvae of the barnacle Balanus amphitrite [ 8 ]. Two other chlorinated metabolites, 4-chloro-1-hydroxy-3-methoxy-6-methyl-8-methoxycarbonyl-xanthen-9-one and chloroisosulochrin dehydrate were isolated from the extract of the endophytic fungus Penicillium citrinum HL-5126 from the mangrove Bruguiera sexangula ; however, these revealed low antibacterial activity with MIC values of 50 μM [ 9 ].…”

“… Moreover, xanthone structures have recently been identified as lichen or fungal metabolites. In the plant kingdom, 24 families have been reported so far to contain xanthones, while extensive studies on xanthone chemistry were made, e.g ., in the Clusiaceae , Gentianaceae , Guttiferae and Hypericaceae families, etc . Because of interesting biological and pharmacological features, such as antioxidant, antimicrobial, antiviral, cytotoxic, anticancer, anti‐inflammatory, hepatoprotective and MAO inhibitor activities, etc ., this compound class is attracting growing attention.…”
