New approaches in the pharmacological treatment of obesity

Abstract: Many new substances are currently being investigated for their usefulness in the pharmaco-therapy of obesity. Most drugs interfere with monoamine neuro-transmitter (serotonin, noradrenalin, dopamine and histamine) effects and act as an appetite suppressant. Other approaches are to primarily increase thermogenesis (e.g. beta 3-adrenoceptor agonists), or to decrease fat absorption by inhibiting the pancreatic lipase (orlistat). New promising agents are substances that increase the effect of corticotropin releasi… Show more

“…35) The present investigation using HFD rats shows that FESR extract (400 and 800 mg/kg) can significantly elevate plasma HDL level and reduce LDL, TC, TG and TL levels without significantly influencing the lipid profile of normolipidemic rats. Observations of changes in body weight and daily food intake suggest that HFD+FESR treatment induced no significant changes in body weight throughout the period of study but significantly decrease the daily food intake.…”

“…Meyer (Japanese name; Chikusetsuninjin), have been used as a folk medicine for hypertension, arteriosclerosis and diabetes. 75 In vitro total chikusetsusaponins as well as chikusetsusaponin III, 28-deglucosyl-chikusetsusaponin IV and 28-deglucosylchikusetsusaponin vs inhibit pancreatic lipase activity at concentrations of 125 to 500 ÎŒg/mL. 76 One study showed that the administration of total chikusetsusaponins (1% to 3% in diet) significantly suppressed weight gain in DIO mice.…”

“…However, diet and physical exercise are usually not very effective, and most patients regain weight even after successful weight loss [1]. Obesity is a major social health problem in many developed countries, and a risk factor associated with the development of type II diabetes, coronary heart disease, hyperlipidemia, hypertension and metabolic syndromes [2,3], certain types of cancer and osteoarthritis [4]. In recent years, several anti-obesity drugs have been approved and marketed to treat obesity.…”
