New application of high pressure reversed‐phase liquid chromatography in lipids

Abstract: High pressure reverse-phase liquid chromatography has been used to separate saturated fatty acids, their methyl esters, polyunsaturated fatty acids, and triglycerides. Rapid separation of fatty acids differing in chain length and number of double bonds has been accomplished. Analysis time was less than 10 min in most cases. The high pressure reverse chromatography resulted in better separations of polyenoic acids than can be accomplished by conventional argentation silicic acid column chromatography. The analy… Show more

“…Also, separated fatty acids could not be recovered from the column. Recently, reverse-phase HPLC has facilitated excellent separation of simple fatty acids or a methyl ester mixture (2)(3)(4), but the strength of absorbance (below 205 nm) of fatty acids depends on the number of double bonds in the fatty acids, and quantitation of each fatty acid requires the collection and gas liquid chromatographic analysis of all HPLC separated compounds. The low sensitivity and complexity of the quantitation of each fatty acid have led to the use of aromatic derivatives which have specific and strong absorbance, such *To whom correspondence should be addressed.…”

“…Long-chained unsaturated fatty acid methylesters can be separated by high-pressure liquid chromatography on silica gel impregnated with silver nitrate [15]. But isocratic reversedphase HPLC separations of the methylesters of saturated and unsaturated fatty acids are known [16,17]; it i s possible to refine such separations with the presently available "reversed-phase" column fillers even further, and this paper deals with a number of such examples. Numerous separation examples [18-211 show that analytical HPLC separations of esters may also be effected with similar results in preparative quantities.…”

“…Hace casi un cuarto de siglo, Pei et al, (1975) publicaron el primer trabajo en el que se proponía un método de HPLC para la separación de TGs de cadena media en una columna de fase reversa. Desde ese momento, otros investigadores han continuado la mejora de los métodos para la separación de TGs.…”
