Neutroprotective efficacy of sodium tanshinone B on hippocampus neuron in a rat model of focal cerebral ischemia

Abstract: Preconditioning with STB appears to be a simple and promising strategy to reduce or even prevent cerebral l/R injury and has potential for future clinical application.

“…A 0.24 mm diameter nylon filament (tip diameter 0.32 ± 0.02 mm; Beijing Sunbio Biotech, Beijing, China) was inserted into the ICA through the ECA stump and was gently advanced (10 mm) to occlude the origin of the middle cerebral artery. After 1.5 h of MCAO, the filament was removed to restore blood flow (reperfusion) [19]. The rats were then allowed to recover after an incision closure and were housed individually.…”

“…For example, TCM preparations such as Huanshaodan decoction 124 and Tianma Cuzhi granules 125 were found to reduce the N -methyl- d -aspartic acid (NMDA) receptor activity in both cerebral cortex and hippocampus. Single active components, such as sodium tanshione B 45 and senegenin 44 decreased NMDA receptor protein expression and mRNA levels of its NR2B subunit in hippocampus. The classical formula Buyang Huanwu decoction 126 decreased neurological deficit scores via down-regulating AMPA receptors, glutamate receptor-1 RNA and glutamate receptor-2 protein expression.…”

“…Using Evan׳s blue dye extravasation and/or brain water content assessment, large doses of TCM preparation, Chinese materia medica, and active compounds were demonstrated to restrain BBB disruption and brain tissue edema after I/R, although the mechanisms for the effects of each TCM may differ. For example, combinations of total alkaloids from Gouteng (Ramulus Uncariae cum Uncis) and Xiatianwu (Rhizoma Corydalis Decumbentis) 101 and the active compound sodium tanshinone B 45 were thought to act through anti-oxidation, whereas the TCM preparation Naomaitong preparation 68 and Guizhi Fuling capsules 67 may produce beneficial effects by anti-inflammatory mechanisms. In contrast, FBD formula 70 and Chinese materia medica Zhimu (Rhizoma Anemarrhenae) 78 may act through inhibition of leukocyte infiltration, whereas the TCM preparation Shexiang Xingnaoning preparation 102 as suggested may act by anti-thrombotic mechanisms.…”

“…Tanshinone IIB significantly reduced the focal infarct volume, cerebral histological damage and apoptosis in rats subjected to MCAO [97]. After tanshinone treatment of rats subjected to MCAO, the levels of glutamate were significantly lower in the hippocampus, and NMDAR1 optical density was reduced in the CA1 region [98].…”
