Neurodegeneration in multiple sclerosis: novel treatment strategies

Abstract: In recent years it has become clear that the neuronal compartment already plays an important role early in the pathology of multiple sclerosis (MS). Neuronal injury in the course of chronic neuroinflammation is a key factor in determining long-term disability in patients. Viewing MS as both inflammatory and neurodegenerative has major implications for therapy, with CNS protection and repair needed in addition to controlling inflammation. Here, the authors' review recently elucidated molecular insights into inf… Show more

“…32,38 Además, en cultivos se ha observado que las células T CD8+ inducen muerte neuronal. 39,40 Estas observaciones sugieren que las células CD8+ también participan en la patología de la EM. 35 Las células T CD8+ interactúan con células que expresan el complejo mayor de histocompatibilidad de clase I (MHCI), el cual se manifiesta en todas las células nucleadas, 32 formando una sinapsis inmunológica estabilizada por las moléculas de adhesión LFA-1 e ICAM-1.…”

“…In susceptible individuals, exposing to environmental factors induces neurodegenerative autoimmune response against CNS, leading to inflammation, demyelination and neuronal injury [7]. Given the critical increase in the rate of the disease over the past two decades and considering the point that rearrangement of genetic materials cannot be responsible for such fast growth, it becomes plausible that changes in environmental factors such as nutrition and life style could have contributed to the rise in disease rates [6].…”

“…This is an accepted metric in common use amongst neuro-radiologists and neurologists in diagnostic and monitoring applications in management of progressive MS patients. 12 There is a general reluctance for some neurological specialists to order repetitive MRI due to the limitation of therapeutic alternatives other than diet, physical therapy, and medications. Unfortunately, many of the available medications have very undesirable side effects, and often prove of limited efficacy.…”
