Nephroprotective effect of Apium graveolens L. against Cisplatin-induced nephrotoxicity

Abstract: Background Cisplatin is extensively used in treating cancers, and its primary side-effect is nephrotoxicity. It accumulates in proximal convoluted tubules where it promotes cellular damage by oxidative stress, apoptosis, and inflammation, etc. In Unani medicine, Tukhm-e - Karafs (Apium graveolens L.) (TK) is mentioned in the literature to manage various kidney ailments due to its diuretic and deobstruent activities. … Show more

“…SIRT1 is a NAD(+)-dependent protein deacetylase expressed primarily on renal tubular cells and podocytes, where it modulates many signaling pathways and promotes cell survival. It deacetylates critical metabolic players, boosting SOD and CAT activity, and it can also decrease inflammation by inhibiting NF-κB activity via deacetylation of the p65 subunit at Lys310 [1,[30][31][32]. It is believed to be a cellular cytoprotective signal that promotes the nuclear translocation/transcriptional activity of Nrf2, resulting in the activation of downstream antioxidant signals such as HO-1.…”

“…It is widely accepted as a representative kidney disease model to investigate acute nephrotoxicity [ 27 , 28 ]. Numerous studies have verified that when an acute renal injury occurs, blood BUN, serum creatinine, and glomerular filtration rate are increased in cisplatin-exposed rodents [ 29 , 30 , 31 ]. Furthermore, it has been established that cisplatin can induce histological changes including mononuclear cell infiltration, hemorrhages, tubular dilation, and vacuolation in kidney tissues [ 30 , 32 ].…”

“…Each blood sample obtained from each mouse was collected into a plain sample vial with a capacity of 4 mL that was clearly labeled. 19 The serum was separated by centrifugation at 11,000 rpm for 10 minutes to measure the total cholesterol level. 10 μL of serum and standard 1 mL of reagent each were added and incubated for 10 min at room temperature.…”
