Neoplastic extracellular matrix environment promotes cancer invasion in vitro

Abstract: The invasion of carcinoma cells is a crucial feature in carcinogenesis. The penetration efficiency not only depends on the cancer cells, but also on the composition of the tumor microenvironment. Our group has developed a 3D invasion assay based on human uterine leiomyoma tissue. Here we tested whether human, porcine, mouse or rat hearts as well as porcine tongue tissues could be similarly used to study carcinoma cell invasion in vitro. Three invasive human oral tongue squamous cell carcinoma (HSC-3, SCC-25 an… Show more

“…However, such protein mixtures lack the complex 3D architecture which also influences proteolytic actions during cell invasion [3]. The described organotypic leiomyoma 3D solid disc model utilizes human tissue material with authentic components of the tumour microenvironment (TME) such as fibroblasts, insoluble and soluble ECM components, and a complex tumour tissue architecture including nerves and vessels [3,4,5]. Although cells in the myoma discs are not viable, their presence is important as they simulate the in vivo tumour cellular environment [6].…”

“…The hypoxic myoma discs mimic the TME of solid human tumours, and they contain a variety of non-vital cells and soluble invasion-related factors such as lysyl oxidase [16]. Recently, we showed that the invasion efficiency of the OTSCC cells into myoma depends on the amount of tenascin-C and soluble growth factors and their receptors in the tissue discs [17]. Myogel, on the other hand, provides a soluble human TME matrix for cancer studies.…”

“…ECM remodelling is generally thought to promote invasive tumour phenotypes, including oral tongue cancer [ 53 55 ]. Secreted tumour EVs coated with fibronectin promote nascent adhesion assembly and increase cell motility [ 56 ].…”
