Nature-Inspired Bioactive Compounds: A Promising Approach for Ferroptosis-Linked Human Diseases?

Abstract: Ferroptosis is a type of cell death driven by iron overload and lipid peroxidation. It is considered a key mechanism in the development of various diseases such as atherosclerosis, Alzheimer, diabetes, cancer, and renal failure. The redox status of cells, such as the balance between intracellular oxidants (lipid peroxides, reactive oxygen species, free iron ions) and antioxidants (glutathione, glutathione Peroxidase 4), plays a major role in ferroptosis regulation and constitutes its principal biomarkers. Ther… Show more

“…Edible mushrooms contain the ferroptosis inhibitor gallic acid, a natural hydroxybenzoic acid, which is also present in various foods such as nuts, red fruits, olive oil, green tea, and vegetables [ 229 ]. Ferroptocide is an inhibitor of thioredoxin, a central redox system in mammalian cells, that induces ferroptosis, which is the unique form of programmed death distinct from apoptosis, and maintains redox balances in sickle red blood cells [ 230 , 231 , 232 ].…”

“…Recent studies have identified several natural compounds with ferroptosis inhibitory properties ( Soriano-Castell et al, 2021 ). These compounds target various molecular pathways involved in ferroptosis, including lipid peroxidation, iron metabolism, glutathione utilization, and antioxidant defense ( El Hajj et al, 2023 ). Among these compounds, polyphenols, flavonoids, and terpenoids have emerged as potential therapeutic agents for the treatment of fibrosis ( Wu et al, 2022 ; Yang et al, 2022 ; El-Horany et al, 2023 ; Wang et al, 2023 ; Zhu et al, 2023 ).…”
