Enhanced drug efflux through ATP-binding cassette transporters, particularly P-glycoprotein (P-gp), is a key mechanism underlying multidrug resistance (MDR). In the present study, we investigated the inhibitory effects of pinostrobin and tectochrysin on P-gp in MDR cancer cells and the underlying mechanisms. Fluorescence substrate efflux assays, multidrug resistance 1 (MDR1) shift assays, P-gp ATPase activity assays, Western blotting, and docking simulation were performed. The potential of the test compounds for MDR reversal and the associated molecular mechanisms were investigated through cell viability assay, cell cycle analysis, apoptosis assay, and further determining the combination index. Results demonstrated that pinostrobin and tectochrysin were not the substrates of P-gp, nor did they affect the expression of this transporter. Both compounds noncompetitively inhibited the efflux of rhodamine 123 and doxorubicin through P-gp. Furthermore, they resensitized MDR cancer cells to chemotherapeutic drugs, such as vincristine, paclitaxel, and docetaxel; thus, they exhibited strong MDR reversal effects. Our findings indicate that pinostrobin and tectochrysin are effective P-gp inhibitors and promising candidates for resensitizing MDR cancer cells.

Increased resistance of Enterococcus faecium, Staphylococcus aureus, Klebsiella pneumoniae, Acinetobacter baumannii, Pseudomonas aeruginosa, and Enterobacter sp. (ESKAPE) pathogens against various drugs has enhanced the urge for the development of alternate therapeutics. Quorum sensing (QS) is a density dependent cell-to-cell communication mechanism responsible for controlling pathogenicity with the regulation of gene expression. Thus, QS is considered a potential target for the development of newer anti-biofilm agents that do not depend on the utilization of antibiotics. Compounds with anti-QS effects are known as QS inhibitors (QSIs), and they can inhibit the QS mechanism that forms the major form in the development of bacterial pathogenesis. A diverse array of natural compounds provides a plethora of anti-QS effects. Over recent years, these natural compounds have gained importance as new strategies for combating the ESKAPE pathogens and inhibiting the genes involved in QS. Different pharmacognostical and pharmacological studies have been carried out so far for identification of novel drugs or for the discovery of their unique structures that may help in developing more effective anti-biofilm therapies. The main objective of this review is to discuss the various natural compounds, so far identified and their employed mechanisms in hindering the genes responsible for QS leading to bacterial pathogenesis.

Introduction: Medicinal plants have been investigated for possible anti-cancer effects. One of them is Gnetum gnemon L (melinjo). This study aims determined in vitro antioxidant activity and the cytotoxic effects of polar, semipolar and non polar melinjo seed fractions againts HeLa cell line. Methods: The melinjo seed were extracted with ethanol as a solvent. Then, the fractionation was done using liquidliquid extraction method with three different polarity solvent. Cytotoxic activity was carried out using 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) colorimetric assay in HeLa cell lines at concentrations ranging from 25 to 400 μg/mL. Antioxidant activity was determined by the diphenyl picryl hidrazil (DPPH) radical scavenging method. Results: Phytochemical screening indicated the presence of phyto-constituents like flavonoids, terpenoid and tanin. The DPPH scavenging activity by the melinjo seed aqueous, ethyl acetate and N-hexane fraction was 733,12 ± 18,95 μg/mL; 68,40 ± 1,9 μg/mL and 2035,70 ± 65,59 μg/mL, respectively. The cytotoxic activity of the melinjo seed fractions showed that the ethyl acetate was the most active fraction against HeLa cell line with IC 50 value 45,27 μg/mL. Conclusion: In this study, we have observed that the melinjo seed fractions exhibited antioxidant and cytotoxic activity against HeLa cell lines. This is presumably due to the content of phytochemicals and stilbenoids such as resveratrol and gnetin C. Melinjo seeds are more potent as anticancer Compared with other plants that also contain RSV such as grape extract (Vitis vinifera L.) against lung cancer cells (A549). From the three fractions, the ethyl acetate fraction had the highest antioxidant and cytotoxic effect compared to the water and n-hexane fractions. Gnetum gnemon L. can be considered as a potential source of anticancer agents. However, more research is needed to determine the mechanism of action.
