Natural Vaccinia Virus Infection: Diagnosis, Isolation, and Characterization

Abstract: Natural infections of Vaccinia virus (VACV)-the prototype species of the Orthopoxvirus genus, from the family Poxviridae and subfamily Chordopoxvirinae-cause an occupational emergent zoonotic disease that is primarily associated with the handling of infected dairy cattle. In humans, VACV infection is characterized by skin lesions, primarily on the hands, and accompanied by systemic symptoms such as fever, myalgia, headache, and lymphadenopathy. The diagnosis of VACV is usually performed according to the method… Show more

“…ELISA was performed for rodent blood samples following a protocol also described previously ( 28 ). For each plate, 1 positive control (serum from Mus musculus experimentally infected with VACV-Guarani P1) ( 29 ) and 3 negative controls (serum of noninfected M. musculus ) were added.…”

“…The plaque-reduction neutralization test (PRNT) protocol was used as described by Kroon et al, (2016). The PRNT was conducted in Vero cells, and the VACV-WR strain purified on a sucrose gradient was used as the control virus.…”

“…To detect VACV DNA we used real-time PCR 55 targeting the C11R and A56R genes, and semi-nested PCR targeting C11R and A26L genes [19]. A 56 molecular approach was performed in serum and tissue samples from rodents and marsupials, and …”
