Nardosinone protects H9c2 cardiac cells from angiotensin II-induced hypertrophy

Abstract: Pathological cardiac hypertrophy induced by angiotensin II (AngII) can subsequently give rise to heart failure, a leading cause of mortality. Nardosinone is a pharmacologically active compound extracted from the roots of Nardostachys chinensis, a well-known traditional Chinese medicine. In order to investigate the effects of nardosinone on AngII-induced cardiac cell hypertrophy and the related mechanisms, the myoblast cell line H9c2, derived from embryonic rat heart, was treated with nardosinone (25, 50, 100, … Show more

“…It was found that, in murine MI/R injury models, the mRNA level of Ang II markedly increased in hearts on day 28, but VCP979 treatment reversed this elevation. consistent with previous reports (41,46), the addition of Ang II induced significant collagen synthesis in NcFs and hypertrophy in NcMs. The present results further revealed that the addition of VcP979 prevented NcFs from Ang II-induced collagen synthesis and NcMs from Ang II-induced hypertrophy in the cell culture system.…”

“…The hypertrophic response of H9c2 cells was induced by angiotensin II (1µmol/L) incubating for 24h [20]. Neonatal rats’ ventricular myocytes (NRVM) from 1-2 days old Sprague-Dawley rats were cultured as described previously [21, 22].…”

“…Since increased oxidative stress is associated with arrhythmogenesis, this may act to suppress arrhythmias (1). Nardosinone, one of the major extracts from Nardostachys, has been shown to prevent angiotensin II (AngII)-induced cardiac hypertrophy in embryonic rat hearts, particularly at a concentration of ≥100 µmol/l by inhibiting the phosphatidylinositol 3-kinase (PI3K)/Akt and MEK/ERK pathways (23).…”
