Nanophotonic Sialidase Immunoassay for Bacterial Vaginosis Diagnosis

Abstract: Bacterial vaginosis (BV) affects reproductive-age women and can lead to pelvic inflammatory disease, postpartum endometritis, and preterm labor/delivery and predisposes the infection of sexually transmitted diseases. Typically, BV diagnosis involves the analysis of vaginal swab samples via microscopy operated by highly skilled personnel. Hence, novel approaches for BV diagnosis are an existing need. In response, the first immunosensing platform targeting sialidase, a BV biomarker, is reported. The nanophotonic… Show more

“…For example, real-time photoluminescent biosensing systems targeting protein/enzyme biomarkers and pathogens have been previously developed by using GO-coated microwells. In particular, these platforms employ fluorescence-tagged antibodies as bioprobes which can be quenched in the absence of the analyte. ,, However, as far as we are concerned, recombinant proteins labeled with fluorophores have not been demonstrated to operate as photoluminescent probes targeting antibodies when incubated in GO-coated surfaces. Apart from offering an advantageous platform for the facile determination of COVID-19 seroconversion (circumventing the employment of two bioreagents with capture and detection purposes, blocking and washing steps), this research sheds light in the aforementioned gap in the literature.…”

“…A score is then assigned to these observations accordingly: 0−3 (normal flora), 4−6 (intermediate or mixed flora), and 7−10 (BV). 11,12 Figure 3A shows the analytical behavior of the analyzed clinical samples. In general, the samples from NM patients exhibited a concentration lower than 25.1 ng mL −1 , and the samples from BV patients displayed a concentration higher than 25.1 ng mL −1 , which is consistent with an immunoassay reported previously.…”

“…Therefore, SLD can be employed as a biomarker of BV. 11,12 A common technique for the detection of SLD in vaginal swabs is based on the enzymatic hydrolysis of methoxyphenyl acetyl muramic acid via the catalytic character of SLD, thereby initiating the formation of methoxyphenol, which is then measured. Usually, in this technique, methoxyphenol concentrations greater than 5.1 nM are considered as BV positive.…”
