Nanoparticles for targeted cancer radiotherapy

Abstract: Radiotherapy, where ionizing radiation is locally delivered either through an external beam or by surgically implanting radionuclidebased seeds in the tumor, is one of the gold standard treatments for cancer. Due to the non-selective nature of radiation, healthy tissue surrounding the cancerous region is usually affected by the treatment. Hence, new strategies, including targeted alpha therapy, are being studied to improve the selectivity of the treatment and minimize side effects. Several challenges, however,… Show more

“…Radiotherapy (RT) is one of the most widely used and effective treatments for malignant solid tumors in clinical settings (Citrin, 2017 ; Pallares & Abergel, 2020 ; Lehrer et al., 2021 ). The anticancer effect of RT is based on DNA damage of cancer cells via high-energy photons or charged particles (Meidanchi et al., 2015 ; Jiang et al., 2018a , 2018b ; Hahn et al., 2021 ).…”

“…There was no significant difference in toxicity between the two groups and no treatment-related death occurred ( 278 ). Although this is very promising data in the sarcoma field where very few patients achieve pathologic complete response with preoperative radiation and possibly in other cancers as well, a lot of challenges lie ahead for the clinical implementation of this technology and overcoming its limitations, particularly optimization of delivery ( 279 ).…”

“…[ 4–8 ] As one of the RT modalities, brachytherapy exhibits prominent merits of precisely localized radiation with high efficient tumor destruction and less normal tissue damage. [ 9–11 ] In brachytherapy, radioisotope sources with short‐range are directly placed within or around the tumor lesions. The precise, continuous, and closed tumor irradiation can generate reactive oxygen radicals and induce DNA double strands breakage, and this is the main cause of tumor regression.…”
