Nanoparticle Superstructures Made by Polymerase Chain Reaction: Collective Interactions of Nanoparticles and a New Principle for Chiral Materials

Abstract: Polymerase chain reaction (PCR) was realized on the surface of gold nanoparticles (NPs) as a tool for self-organization at nanoscale and as a step toward programmable production of sufficient quantities of functional metallic superstructures. The assembly is controlled by varying the density of the primer on the surface of gold NPs and the number of PCR cycles generating a mixture of dimers, trimers, tetramers, etc., with gradually increasing complexity. This process leads to strong chirality of the assemblies… Show more

“…These researchers also reported that chiral pyramidal structures were produced using four types of AuNPs of different sizes. Similar chiral AuNP assemblies have been independently prepared by Chen et al 21 Kim et al 22 also reported on pyramidal or octahedral structures created with AuNPs and DNA, in which the AuNPs were located at the vertices of the polyhedrons. These researchers called the method 'aqueous-phase anisotropic sequential ligand replacement strategy' , and it could attach DNA strands up to six times at right angles to each other on an AuNP (Figure 2e).…”

“…In this way, molecules like oligonucleotides, peptides, or PEG can be readily linked to GNPs. [18][19][20] The advantage of physical adsorption is that the nanoparticles have a minimum effect on the structure and function of the biological molecule. With native structure intact, the activity, selectivity, and specificity of the biological molecule towards a particular target are largely unaffected.…”

“…This can give an exquisite level of control, as the polymerase chain reaction can be performed directly on the surface of GNPs for this kind of organization. 18 Any or all of these approaches may potentially be modified for gene or RNA stabilization and delivery as well, though remain to be tested. The possibilities for functionalization upon GNPs are vast.…”

“…In addition to these well-established applications, a new area that is emerging is plasmonics (Figure 3) 62,63 and especially chiroplasmonic assemblies and composites 64,65 . DNA-bridged NP assemblies can have a particularly essential role in the latter because they afford automatic assembly of such superstructres with exceptionally strong polarization rotation using PCR technique.…”
