Nanoparticle delivery systems for substance use disorder

Abstract: Innovative breakthroughs in nanotechnology are having a substantial impact in healthcare, especially for brain diseases where effective therapeutic delivery systems are desperately needed. Nanoparticle delivery systems offer an unmatched ability of not only conveying a diverse array of diagnostic and therapeutic agents across complex biological barriers, but also possess the ability to transport payloads to targeted cell types over a sustained period. In substance use disorder (SUD), many therapeutic targets h… Show more

“…From a translational perspective, nanotechnology-based drug delivery is employed in the present work to improve the therapeutic index of BAPN due to the associated cardiotoxicity 49 and neurotoxicity 50 . There are hitherto three main categories of nanoparticles, namely organic, inorganic and hybrid based on the chemical properties 67 . Among these nanomaterials, inorganic nanoparticles offer some advantages for the implementation of targeted drug delivery, notably in bioavailability.…”

“…Another substance for brain therapy applications is inorganic-based nanostructures, such as iron, gold, and silica. These well-formulated nanoparticles have shown inherent and unique magnetic, optical, electrical, and physical properties that could be utilized in targeted drug delivery and imaging applications ( Mitchell et al, 2021 ; Kasina et al, 2022 ; Khademolqorani and Banitaba, 2022 ).…”

“…Although highly charged AuNPs can activate an immune response in the brain, surface modifications can be added to help alter the charge ( Kus-Liskiewicz et al, 2021 ). AuNPs usually consist of an inner inorganic layer that is surrounded by a nonpolar organic layer that helps stabilize the core and attaches to the NBT ( Kasina et al, 2022 ). Surface modifications can be added to aid with targeting, while the small size of AuNPs helps with crossing the BBB.…”
