“…The RBD-loaded nanoparticles were prepared by ionotropic gelation as previously described [ 23 ]. Briefly, a sodium tripolyphosphate (TPP, 0.167 mg/mL) solution mixed with RBD protein (0.3 mg/mL) was added dropwise into a TMC solution in a HEPES buffer (pH 7.4) containing 1% ( v/v ) Tween 80 under continuous stirring for 1 h at room temperature.…”

“…The supernatant was harvested for estimating the amount of unbound RBD by Pierce™ BCA Protein Assay Kit (Thermo Fisher Scientific, Rockford, IL, USA). The efficiency of protein loading in TMC NPs was calculated as described previously [ 23 ]. To characterize the physical properties of NPs, the mean of particle size, polydispersity index (PDI) and the zeta potential were evaluated by a zetasizer (Malvern Instruments Ltd., Malvern, UK).…”

“…Recently, Jearanaiwitayakul et al prepared nanovaccine composed of non-structural protein (NS1) of dengue virus (PENV) vaccine and TMC by the ionic gelation method to protect dengue virus as the most common mosquito-borne viral disease [ 45 ]. The nanovaccine potentially stimulated monocyte-derived DCs (MoDCs) and resulted in increased expression of CD83 as the maturation marker, and CD80 and CD86 as costimulating molecules, and marked secretion of innate immune cytokines.…”

“…VLPs based DENV vaccines are described by Zhang et al, 2020, but none of them were developed with NS1 proteins [53]. One example is given with C-terminal truncated DENV-2 NS1 loaded in N,N,N, trimethyl chitosan nanoparticles (NS11-279TMC NPs) investigated in murine model and in human ex vivo [40]. In human ex vivo model, it has been demonstrated that TMC particles deliver NS11-279 protein in monocyte-derived dendritic cells (MODCs) and also stimulate those cells resulting in an increase expression of maturation marker (CD83), costimulatory molecules (CD80, CD86 and HLA-DR) and secreting diverse immune cytokines/chemokines [40].…”

“…Immunization with NS11-279TMC NPs resulted in both B cell and T cell responses leading to IgG production and CD8+ T cells activation. One important finding is that DENV2 NS11-279directed antibodies have ability to kill DENV-infected cells through antibody dependent complement-mediated cytotoxicity [40]. Consequently, a such NS1-based vaccine candidate is of great interest in relation to the properties of TMC as a suitable adjuvant which enhances delivery and promote immunogenicity of viral antigen.…”
