This study investigated the effects of dietary phosphatidylcholine (PC) at different growth stages of Litopenaeus vannamei (L. vannamei) with an initial weight of 0.70 ± 0.00 g (juvenile) and 4.25 ± 0.00 g (subadult) for 8 weeks. Six isonitrogenous and isoenergetic diets containing PC (0, 1.7, 5.1, 6.8, 10.2 and 13.6 g/kg) were confected using formulated with redfish meal, dehulled soybean meal and peanut meal as protein source, fish oil and corn oil as fat source. The juveniles were reared in 0.3 m3 aquarium at the density of 40 in triplicates, as well as the subadults in 0.5 m3 at the density of 40 in triplicates. Results showed PC level had significant effects on weight gain rate (WGR), specific growth rate, feed conversion ratio in juvenile and subadult shrimps and hepatosomatic index in juvenile shrimps (p < 0.05). No significant differences were found in moisture, whole body protein and crude ash (p > 0.05). Crude fat of juveniles had no significant change (p > 0.05), but subadults increased significantly (p < 0.05). Both juveniles and subadults showed that triglyceride and low‐density lipoprotein presented a downward trend with PC elevating, but high‐density lipoprotein increased gradually. Cholesterol in juveniles presented upward, but downward in subadults (p < 0.05). Dietary PC also significantly reduced the aspartate transaminase in juveniles and alanine transaminase in subadults (p < 0.05). PC supplementation significantly improved the glutathione S‐transferase activity and reduced the content of malondialdehyde in the hepatopancreas (p < 0.05). PC supplementation significantly improved gill filament sodium‐potassium adenosine triphosphatase enzyme of juvenile shrimps (p < 0.05), but subadult shrimps were not influenced (p > 0.05). PC supplementation significantly elevated the content of hepatopancreas eicosapentaenoic acid, highly unsaturated fatty acid (HUFA) and n‐3HUFA (p < 0.05), but the content of linoleic acid significantly reduced (p < 0.05). The broken‐line and quadratic regression model based on WGR showed the optimum dietary PC requirement was 10.3 g/kg and 6.5 g/kg for juveniles and subadults, respectively.

In the field, moulting and salinity drop in the water due to excessive rainfall have been mentioned to be risk factors for WSSV outbreaks. Therefore, in this study, the effect of an acute change in environmental salinity and shedding of the old cuticle shell on the susceptibility of Penaeus vannamei to WSSV was evaluated by immersion challenge. For testing the effect of abrupt salinity stress, early premoult shrimp that were acclimated to 35 g L were subjected to salinities of 50 g L , 35 g L , 20 g L , 10 g L and 7 g L or 5 g L and simultaneously exposed to 10 SID mL of WSSV for 5 h, after which the salinity was brought back to 35 g L . Shrimp that were transferred from 35 g L to 50 g L , 35 g L and 20 g L did not become infected with WSSV. Shrimp became infected with WSSV after an acute salinity drop from 35 g L to 10 g L and lower. The mortality in shrimp, subjected to a salinity change to 10 g L , 7 g L and 5 g L , was 6.7%, 46.7% and 53.3%, respectively (P < 0.05) For testing the effect of moulting, shrimp in early premoult, moulting and post-moult were immersed in sea water containing 10 SID mL of WSSV. The resulting mortality due to WSSV infection in shrimp inoculated during early premoult (0%), ecdysis (53.3%) and post-moult (26.72%) demonstrated that a significant difference exists in susceptibility of shrimp during the short moulting process (P < 0.05). The findings of this study indicate that during a drop in environmental salinity lower than 10 g L and ecdysis, shrimp are at risk for a WSSV infection. These findings have important implications for WSSV control measures.

BackgroundSea cucumbers (Apostichopus japonicus) are an imperiled fauna exposed to a variety of environmental condition such as salinity and studies are urgently needed to assess their effects to guide aquaculture efforts. The effects of acute salinity stress on coelomic fluid osmotic pressure, ion concentrations, the activity of Na+/K+-ATPase in respiratory trees and the histological variations were measured to evaluate the salinity tolerance of sea cucumbers.ResultsSignificant correlations in osmotic pressure were observed between coelomic fluid and ambient environmental salinity. In coelomic fluid, Na+ concentration was observed fluctuated during salinity 18 psu and the inflection point presented at the 6 h. The Na+/K+-ATPase activity in respiratory trees indicated the “U-shaped” fluctuant change and the change trend was opposite with the Na+ concentration. The ions (K+, Cl−) concentration decreased and showed the same tendency at salinity 40 psu with salinity 18 psu. The total coelomocytes counts and phagocytosis of coelomic fluid Na+/K+-ATPase activity indicated fluctuating changes under different salinity stress. Histological variation revealed a negative relation between decreasing salt concentration and tissue integrity. Tissue damages were significantly observed in intestines, muscles and tube feet under low salinity environment (18, 23 and 27 psu). The connective tissue in intestines of A. japonicus exposed to 18 and 23 psu damaged and partly separated from the mucosal epithelium. The significant variations occurred in tube feet, which presented the swelling in connective tissue and a fracture in longitudinal muscles under low salinity (18 psu). The morphological change of tube feet showed the shrinkage of connective tissue under high salinity (40 psu). The amount of infusoria in the respiratory trees decreased or even disappeared in salinity treatment groups (18 and 23 psu).ConclusionThe results inferred that osmoconformity and ionoregulation were seen in sea cucumbers, which contributed to understand the salinity regulatory mechanisms of A. japonicus under acute salinity stress.
