Mycobacterium tuberculosis CysA2 is a dual sulfurtransferase with activity against thiosulfate and 3-mercaptopyruvate and interacts with mammalian cells

Abstract: Cyanide is a toxic compound that is converted to the non-toxic thiocyanate by a rhodanese enzyme. Rhodaneses belong to the family of transferases (sulfurtransferases), which are largely studied. The sulfur donor defines the subfamily of these enzymes as thiosulfate:cyanide sulfurtransferases or rhodaneses (TSTs) or 3-mercaptopyruvate sulfurtransfeases (MSTs). In Mycobacterium tuberculosis, the causative agent of tuberculosis, the gene Rv0815c encodes the protein CysA2, a putative uncharacterized thiosulfate:cy… Show more

“…M. tuberculosis ProX binds polyphenols such as phloretin, which can be used as a carbon source [23]. Mutants of the proXVWZ operon showed problems in accumulating glycine betaine, deficiency in survival and growth in macrophages, and impaired the bacillus growth in a medium with a high osmolarity [57].…”

“…Our RNA-seq analysis identified that Ebselen displayed bactericidal activity in M. bovis BCG through cysA2 and glpD2 down-regulation which are the major regulators in sulfur and glycerol metabolism respectively. CysA2 is an essential regulator in the sulfur assimilation pathway [23] and its dysregulation could impair M. tuberculosis survival in macrophages [24]. In parallel, a previous study demonstrated that inhibition of glycerol metabolism by 2-aminoquinazolinones via glpD2 down-regulation could kill M. tuberculosis in vitro [25].…”

“…For firefighters, they have more chance to breathe or face the air with high concentration VOCs, and researches showed that VOCs could absorb via skin directly [55][56][57]. Cyanide and SO2 were typically toxic for human health, they not only induce oxidative damage in many organisms simultaneously but the toxic effects permanently [58,59]. The SO2 and its derivatives SO32-could promote cancer development and metastasis through integrin αvβ3 activation or other molecules [60,61].…”
