Purpose: Endometriotic cysts are known to transform into ovarian cancers, such as clear cell and endometrioid carcinomas. We hypothesized that an iron-rich environment produced by the repetition of hemorrhage in the endometriotic cysts during the reproductive period may play a crucial role in carcinogenesis in the cysts through the iron-induced persistent oxidative stress. Experimental Design: Contents of human ovarian cysts, including 21 endometriotic cysts, 4 clear cell carcinomas, and 11nonendometriotic cysts, were analyzed for the concentrations of free ''catalytic'' iron, lactose dehydrogenase, potential antioxidant, lipid peroxide, and 8-hydroxy-2 ¶-deoxyguanosine (8-OHdG). Iron deposition and 8-OHdG levels were also analyzed histologically. Reactive oxygen species and the mutagenicity of the contents in endometriotic cyst were determined in vitro. Results: The concentration of free iron in endometriotic cysts (100.9 mmol/L) was significantly higher than that in nonendometriotic cysts (0.075 mmol/L; P < 0.01). The average concentrations of lactose dehydrogenase, potentialantioxidant, lipidperoxide, and 8-OHdG were alsosignificantly higher in endometriotic cysts (P < 0.01).There was a correlation between the concentration of free iron and that of 8-OHdG (P <0.01). Histologically, we could observe iron deposits more abundantly in endometriotic cysts than in nonendometriotic cysts (P < 0.01).The level of 8-OHdG in carcinoma associated with endometriosis was higher than that of carcinoma without endometriosis (P <0.05).

Praziquantel (PZQ) is the drug of choice against schistosomiasis. However, PZQ was found not fully effective. A new trend from natural plant sources was used to compete schistosomiasis. In the present study, the antischistosomal activities of Curcumin extract and /or PZQ were evaluated. Parasitological and histopathological examination of liver has been explored. Moreover, Surface topography of the recovered worms was inspected by SEM. On the other hand, the total protein and DNA fragmentation percentage were estimated in infected and infected treated mice as compared to control group. Treatment of infected CD1 mice with curcumin extract alone didn't show any detectable changes in the parasitological or histological parameters. The infected mice treated with PZQ only, showed a significant decrease in the total worm burden, egg load (52%), with increase in degenerated eggs (58%) in the oogram pattern.However, treating infected CD1 mice with combined therapy (curcumin extract + PZQ) showed extremely significant reduction in female and male worm count (93% & 84%) respectively. Liver tissue egg load decreased by 88%, intestinal egg load decreased by 97%. Combined therapy decreased granuloma count and diameter with significant increase in degenerated egg in the oogram count. Moreover, the studied biomarkers, related to liver function and DNA damage were significantly improved in infected mice treated with curcumin combined with PZQ as compared to either drug alone. . In conclusion, curcumin extract in combination with PZQ has a synergetic effect and could be more promising in controlling schistosomiasis.Copy Right, IJAR, 2016,. All rights reserved.

Infection with schistosomes invokes severe fibrotic granulomatous responses in the liver of the host. Schistosoma mansoni infection induces dramatic fluctuations in Th1 or Th2 cytokine responses systemically; Th1 reactions are provoked in the early phase, whilst Th2 responses become dominant after oviposition begins. In the liver, various unique immune cells distinct from those of conventional immune competent organs or tissues exist, resulting in a unique immunological environment. Recently, we demonstrated that S. mansoni infection induces unique CD4+ T cell populations exhibiting unconventional cytokine profiles in the liver of mice during the period between Th1- and Th2-phases, which we term the transition phase. They produce both IFN-γ and IL-4 or both IFN-γ and IL-13 simultaneously. Moreover, T cells secreting triple cytokines IFN-γ, IL-13 and IL-4 were also induced. We term these cells Multiple Cytokine Producing Hepatic T cells (MCPHT cells). During the transition phase, when MCPHT cells increase, IL-18 secretion was up-regulated in the liver and sera. In S. mansoni-infected IL-18-deficient mice, expansion of MCPHT cells was curtailed. Thus our data suggest that IL-18 produced during S. mansoni infection play a role in the expansion of MCPHT cells.
