Multispectral optical imaging device for in vivo detection of oral neoplasia

Abstract: Abstract.A multispectral digital microscope ͑MDM͒ is designed and constructed as a tool to improve detection of oral neoplasia. The MDM acquires in vivo images of oral tissue in fluorescence, narrowband ͑NB͒ reflectance, and orthogonal polarized reflectance ͑OPR͒ modes, to enable evaluation of lesions that may not exhibit high contrast under standard white light illumination. The device rapidly captures image sequences so that the diagnostic value of each modality can be qualitatively and quantitatively evalua… Show more

“…At the beginning of the repair process, the extracellular matrix consists of 30% type III collagen, resulting in a more fragile matrix. Collagen fi bers are birefringent and therefore appear bright against a dark background when analyzed under a polarized light microscope 9,10,18 . This technique permits the use of collagen fi brils to characterize the different stages of wound healing 9,18 .…”

“…Autofluorescence originates from a variety of fluorophores in the oral cavity, and is sensitive to alterations in both tissue morphology and biochemistry associated with neoplasia [34,35]. Two optical devices, the VELScope(R) (LED Dental, Inc. White Rock, BC, Canada) introduced 3 years ago, and the new Identafi(R) 3000 Ultra (Trimira, LLC, Houston, Texas), take advantage of the fact that, to a certain degree, we all glow.…”

“…For example, time gating can separate photons based on their optical pathlengths, however, this approach requires highly sophisticated and expensive equipment [10][11][12]. In an alternative method, our group and others have developed a simple linear polarization gating to isolate tissue signals originating from shallow depths in tissues from the diffuse background of the underlying stroma using spectroscopic and imaging setups [13][14][15][16][17][18][19][20][21]. In polarization gated spectroscopy and imaging, polarized photons maintain their incident polarization for a particular number of mean free paths (MFP) depending on density of optical scatterers and their properties such as anisotropy factor [22,23].…”
