Multiplex amplification refractory mutation system PCR (ARMS-PCR) provides convenient method for differentiation of canine parvovirus vaccine and field strains

Abstract: Vaccination with canine parvovirus-2 (CPV-2) modified live attenuated vaccine remains an effective control strategy for preventing parvovirus induced enteritis in dogs. Virus shedding is a common phenomenon few days after vaccination, possessing a diagnostic dilemma for accurate differentiation of CPV-2 vaccine and wild type field strains. Though several molecular approaches are available for differentiation, the present study focuses on a simple, rapid, cost-effective differentiating infected from vaccinated … Show more

“…This approach is closer to be achieved if the exponential growing of the genetic databases is considered mainly if it is also taken into consideration that those animals under a chronic or persistent infection can excrete infectious viral particles for a lifetime period [ 166 , 167 ]. Thus, the use of highly sensitive molecular techniques has, nowadays, given rise to an increase in vaccine virus detections and differentiation [ 153 , 154 , 168 ]. In previous years, several research groups have developed some molecular biology-based CSF genetic DIVA systems.…”

“…These methods were developed for use with high throughput techniques but costs and practicability have so far limited the wider application of these technologies. The tetra-primer amplification refractory mutation system PCR (tetra-primer ARMS-PCR) developed by Ye et al has proved to be an economical and effective method for SNP genotyping [25][26][27] . Positioning the two outer primers at different distances from the polymorphic nucleotide, causes the two allele-specific amplicons differ in length, allowing them to be discriminated by gel.…”
