Multiple Effects of Resveratrol on Osteosarcoma Cell Lines

Abstract: Osteosarcoma (OS) is the most common primary bone sarcoma affecting the life of pediatric patients. The clinical treatment faces numerous difficulties, including the adverse effects of chemotherapies, chemoresistance, and recurrences. In this study, the effects of resveratrol (RSV), a natural polyphenol, on OS cell lines were investigated to evaluate its action as an adjuvant therapy to the current chemotherapy regimens. RSV exhibited multiple tumor-suppressing activities on OS cell lines, inducing a series of… Show more

“…28 In addition to this, these substances promote angiogenesis, which promotes tumor development and allows metastatic spread. 29,30 A variety of growth factors have been implicated in the process of angiogenesis. These molecules-VEGF (vascular endothelial growth factor), fibroblast growth factor-2, and PDGF (platelet-derived growth factor)-are regarded as particularly important.…”

“…A methylation-sensitive restriction endonuclease–PCR (MSRE–PCR) analysis was performed to determine the methylation status of the CpG-rich sites, present in the proximal promotor region of Interleukin-6 (IL-6) and Interleukin-8 (IL-8). Experiments were carried out as described elsewhere [ 15 , 39 , 40 ]. PCR products were analyzed by 2% agarose gel electrophoresis visualized by Gel Red staining (Biotium, Hayward, CA, USA) in a ChemiDoc apparatus (Bio-Rad Laboratories, Hercules, CA, USA), and the image captured in the digital support and densitometric analysis was obtained using the “Image Lab” application (version 5.2.1) of Bio-Rad Laboratories (Hercules, CA, USA).…”

“…However, human epidemiological studies are not having much success in evaluating polyphenols’ effects due to their poor bioavailability and high metabolism rate [ 32 ]. Accumulating data showed in vitro polyphenols anti-proliferative effects on several cancer types, such as prostate [ 33 , 34 ], colon [ 35 , 36 ], breast [ 37 , 38 , 39 ], lung [ 40 , 41 ], bladder [ 42 , 43 ], pancreatic [ 44 , 45 ], leukemia [ 46 , 47 ], osteosarcoma [ 48 , 49 ] and ovarian [ 50 , 51 ], through different mechanisms of action. They can influence important molecular events related to carcinogenesis; these include the modulation of cell cycle signaling, induction of detoxifying and antioxidant enzyme systems, alteration of the epigenome and metabolism, and changes in the expression of key proteins involved in signal transduction pathways (e.g., MAPK and PI3K) or in the activity of transcription factors (e.g., NFkB) [ 52 , 53 , 54 ].…”
