Multifunctional microbubbles for image-guided antivascular endothelial growth factor therapy

Abstract: Abstract. We synthesize multifunctional microbubbles ͑MBs͒ for targeted delivery of antivascular endothelial growth factor ͑antiVEGF͒ therapy with multimodal imaging guidance. Poly-lactic-co-glycolic acid ͑PLGA͒ MBs encapsulating Texas Red dye are fabricated by a modified double-emulsion process. Simultaneous ultrasound and fluorescence imaging are achieved using Texas Red encapsulated MBs. The MBs are conjugated with Avastin, an antiVEGF antibody for treating neovascular age-related macular degeneration ͑AMD͒… Show more

“…PLGA particles may be used to encapsulate absorption and fluorescence dyes in addition to a drug for multimodal imaging using fluorescence (FL), ultrasound (US), or photoacoustic tomography (PAT) [69]. Such multifunctional particles can also be formulated from a component material to reduce side effects of the encapsulated drug [70]. These particles can not only serve as a delivery system for the encapsulated drug but also reduce the harmful side effects through targeted drug delivery.…”

“…The microparticles contain a shell of PLGA, a FDA approved biodegradable polymer material, and a aqueous core that is capable of loading various water solvable drugs, antibodies, and genes. Such multifunctional microparticles can be used for various biomedical applications, including cancer therapy [26] and ophthalmologic disease treatment [27].…”

“…Currently, fluorophores and microbubbles attached with various functional groups have been widely reported in the literature and many of them are available commercially. 39,[46][47][48] Thus, the synthesis of microbubble-based UMF contrast agents become straightforward, which is necessary for bringing this technique into practical application. More importantly, simultaneous multimolecule targeted imaging becomes straightforward by using fluorophores with different excitation and emission wavelengths, which will be investigated in the future.…”
