Multifunctional co-transport carriers based on cyclodextrin assembly for cancer synergistic therapy

Abstract: In the past few years, drug delivery systems have been used extensively to improve solubility, stability, and pharmacokinetics of chemotherapeutic drugs. However, traditional delivery systems fail to fulfill the required standard of effectiveness, primarily owing to issues of drug resistance exhibited by cancer cells and inherent differences among the patients. In this regard, combination therapy offers advantages of synergistic mechanism, reduced drug dosage, and enhanced therapeutic effect, which can be effe… Show more

“…They have a hydrophilic surface on the outside and a lipophilic cavity in the middle. [ 347,348 ] The selective modification of the hydroxyl groups on the outer surface of cyclodextrins can confer a specific function. [ 349 ] The hydrophobic spaces in cyclodextrins can be filled by molecules like azobenzene, ferrocene, adamantane, and their derivatives, ADA.…”

“…By conjugating multiple components, such as different kinds of tumor-targeting ligands, anticancer drugs, or siRNAs, to a single construct, not only can these multifunctional nanosystems target more cancer cells, but also they can achieve effective synergistic therapeutic treatment [ 71 , 72 ]. Investigations of multifunctional therapeutic strategies are progressing rapidly and provide new opportunities in the treatment of cancer [ 73 , 74 ].…”

“…A nanoparticle was constructed to deliver both antisense DNA and siRNA simultaneously [103] . Seven antisense DNA strands were connected to β‐cyclodextrin (a cyclic oligosaccharide composed of seven D‐glucose units), which is very conveniently employed for pharmaceutical applications because of many favorable features [104–107] . In order to bind siRNA to these antisense DNA strands, each strand of the siRNA is extended to the 3’‐direction.…”
