This work presents the construction and application of boron-doped diamond (BDD) thin film electrode as sensor for the determination of three β-agonists, viz. salbutamol, terbutaline and clenbuterol. Although well-known as a chemically inert material, BDD film however shows fouling in detection of these compounds using fixedpotential mode amperometry. A suitable waveform for pulsed amperometric detection (PAD) was developed and used to determine the agonist compounds. It was seen that the developed PAD significantly refreshed the BDD surface for long-term detection in flow injection analysis. Linear working ranges were 0.5-100 µM, 1.0-100 µM and 0.5-50 µM for salbutamol, terbutaline and clenbuterol, respectively. The developed PAD-BDD system was applied to successfully determine salbutamol and terbutaline in commercial pharmaceutical products. The methods were validated with a capillary electrophoresis method.

Ion suppression, a matrix effect that affects quantitative mass spectrometry, is one of the main problems encountered in liquid chromatography/tandem mass spectrometry. Two different clean-up steps for the multi-residue analysis of beta-agonists in urine were evaluated with respect to minimisation of ion suppression, namely, a mixed-phase solid phase extraction (SPE) column, i.e., clean screen Dau (CSD), and a molecular imprinted polymer (MIP) SPE column. Ion suppression experiments revealed that CSD sample clean-up can lead to false negative results for some beta-agonists, and that clean-up using MIP columns is more selective for beta-agonists than the use of CSD columns.

Clenbuterol screening of bovines is done by analysis of urine, for monitoring living animals, and liver, for monitoring animals after slaughter. ELISA has generally been used as the main method for these purposes. Nevertheless, in Europe, methods must be validated according to Commission Decision (EC) 657/2007 criteria, i.e. by use of reference materials. Production of ''in house'' reference materials is a possibility, but the homogeneity, storage temperature, and period of stability of these materials must be investigated in the laboratory itself. This paper reports GC-MS evaluation of an ''in-house''-produced batch of aliquots of bovine urine and liver, fortified with 10.0 ng/ml and 10.0 ng/g clenbuterol, respectively, and stored at -20°C and at -60°C. For urine stored for 20 weeks at -20°C and at 60°C the stability of clenbuterol was proved at the 95% confidence level. For liver, however, it was demonstrated at the same confidence level that clenbuterol was highly unstable during storage for 20 weeks at either of the temperatures studied.
