MT2‐MMP Expression During Early Avian Morphogenesis

Abstract: Membrane-type 2 matrix metalloproteinase (MT2-MMP; also called MMP15) is a membrane-bound protease that degrades extracellular matrix and activates proMMPs such as proMMP-2. MMP-2 expression in avian embryos is well documented, but it is not clear how proMMP-2 is activated during avian embryogenesis. Here, we report that MT2-MMP mRNA is expressed in several tissues including the neural folds and epidermal ectoderm, intermediate mesoderm, pharyngeal arches, limb buds, and dermis. Several, but not all, of these … Show more

“…MMP15 does not seems to be expressed in NC cells per se, but it is expressed in the mesoderm underlying the neural tube and the epidermis in the NC pathways. Furthermore, MMP15 is found in pharyngeal arches and the heart (Patterson, Cavanaugh, Cantemir, Brauer, & Reedy, ). MMP16 is expressed in cranial NC cells during migration (Roth, Kalev‐Altman, Monsonego‐Ornan, & Sela‐Donenfeld, ).…”

“…MMP9 is a secreted protease that localizes to both delaminating and trunk migratory neural crest cells in the chick head and trunk (Monsonego-Ornan et al, 2012), while Xenopus cranial neural crest cells express MMP-14 ( XMT1-MMP ) (Harrison et al, 2004; Tomlinson et al, 2009). MMP15/MT2-MMP , an activator of pro-MMP2, is expressed in the neural folds of the very anterior chick head and open neural plate region in the trunk, while its protein product is observed along the basolateral surface/basement membrane of the neural tube (Patterson, Cavanaugh, Cantemir, Brauer, & Reedy, 2013). In zebrafish, the GPI-linked MMP17b is expressed in crestin -positive cranial neural crest cells, and knockdown of MMP17b function through MOs or chemical inhibitors leads to aberrant migration and patterning of crestin -positive trunk neural crest cells (Leigh et al, 2013).…”

“…For example, mt2-mmp has been reported to be expressed in the pharyngeal arches in both zebrafish and avian embryos (Patterson et al, 2013;Quick et al, 2012). MT2-MMP and, to a lesser extent, MT1-MMP, promote epithelial proliferation during branching morphogenesis in ex vivo mouse submandibular gland cultures.…”
