MPSS profiling of embryonic gonad and primordial germ cells in chicken

Abstract: The massively parallel signature sequencing (MPSS) provides a greater depth of coverage than expressed sequence tag scan or microarray and provides a comprehensive expression profile. We used the MPSS technology to uncover gene expression profiling in the early embryonic gonads and primordial germ cells (PGCs) in the chicken. Total numbers of sequenced signatures were 1,012,533 and 995,676 for the PGCs and gonad, respectively. Using a noise distribution model, we found that 1.67% of all signatures are expresse… Show more

“…During gastrulation, PGCs circulate through the vascular system and settle down in the gonadal anlagen. Such a differentiation pathway, including germ cell lineage during chicken embryo development, is a systematic process, governed by the concerted action of multiple unknown regulatory mechanisms (4)(5)(6).…”

“…4). The absorbance value can be converted into the amine density of each surface by Lambert-Beer law [20,21]. The calculated surface densities of amine groups were 4.61 nm −2 for the APTMS surface, and 2.75 nm −2 for the aminodextran surface.…”

“…To measure the density of the surface amine groups on the aminodextran and APTMS surfaces, UV-visible absorption spectroscopy analysis was used as described in previously published procedures [20,21]. The amine-functionalized surfaces were immersed into 1% 4-nitrobenzaldehyde in anhydrous ethanol under nitrogen atmosphere for 12 h to form the imines.…”

“…The presence of amine and hydroxyl groups results in the reductions of water contact angles of PTFE-PEDA film surfaces from 120°to 45°. The surface densities of amine groups on PTFE-PEDA films are also measured using 4-nitrobenzaldehyde as a testing agent according to the reported method [23,24]. The surface densities of amine groups of the PTFE/PDEA films slightly increase with increases in the plasma powers for EDA plasma polymerization, as values of about 5000 and 5800 groups/nm 2 the measured surface densities of the amine groups for the PTFE-PEDA films are relatively high comparing to the values (around 2-5 amine groups/nm 2 ) reported in literatures [23,24].…”
