Moving Enzyme-Linked ImmunoSorbent Assay to the Point-of-Care Dry-Reagent Strip Biosensors

Abstract: In this work, we described a point-of-care (POC) dry-reagent strip biosensor (DRSB) based on enzyme tracers and portable strip reader for simple, low-cost and sensitive assay of protein detection in minutes. Horseradish Peroxidase (HRP) and Rabbit IgG (R-IgG) were used as a model system for the demonstration of the proof-of-concept. The sandwich-type immunoreactions were performed on the DRSB and the HRP tracers were captured on the test zone of the biosensor. The excess of HRP tracers were captured on the con… Show more

“…In enzyme LFIA the most often used label is horse radish peroxidase (HRP) [3][4][5][6], however alkaline phosphatase [8,9] and cholinesterase [10] were also employed. As far as HRP is concerned, colorimetric [3][4][5][6], chemiluminescent [11,12] and electrochemical [13] detection modes were reported. Substrate solution contained 3,3',5,5'-tetramethylbenzidine (TMB) was predominantly employed for colorimetric detection in HRP-based LFIA [4,6], however 4-chloro-1-naphthol was also used in few studies [5,7].…”

“…Later enzyme immunoassay was realized in conventional LFIA format with specific reagents immobilized in a form of narrow bands [8]. “…shows that AuNPs with a diameter of ~20 nm intrinsically gives a better visibility on paper based strip when accumulated on test line, and many studies [42][43][44] also used this size for similar applications, therefore we used AuNPs with average size around 20 nm as the labels.…”
